摘要
试验旨在探究L-肉碱对乙醇诱导氧化应激损伤的非洲绿猴肾上皮细胞(vero)的保护作用。以vero细胞为研究对象,分别检测不同体积浓度乙醇(0、2.5%、5%、10%、20%)处理vero细胞3 h、L-肉碱(0、0.01、0.1、1、10 mmol/L)作用vero细胞12 h及L-肉碱预处理乙醇诱导氧化损伤的vero细胞12 h后的细胞内超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GPx)活性、总抗氧化能力(T-AOC)水平及丙二醛(MDA)含量。结果表明:与对照组相比,乙醇浓度为5%、10%、20%时,细胞中SOD和GPx活性显著降低(P〈0.05),且MDA水平显著升高(P〈0.05);L-肉碱单独作用细胞时,浓度为1、10 mmol/L组SOD和GPx活性显著高于对照组(P〈0.05);与乙醇处理组相比,1、10 mmol/L的L-肉碱预处理能显著提高乙醇诱导损伤的细胞内SOD和GPx活性及T-AOC水平(P〈0.05),同时使细胞内MDA水平显著降低(P〈0.05)。本试验表明,乙醇能够诱导细胞产生氧化应激,而适宜浓度的L-肉碱可以提高细胞的抗氧化功能;同时验证了1-10 mmol/L L-肉碱对氧化应激所致的vero细胞损伤具有保护作用。
The damage was in protective effect and probable of L-carnitine on vero cells against ethanol-induced oxidative stress vestigated. Using vero cells as research object in vitro culture, the intracellular enzyme activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), and the levels of total antioxidant capacity (T-AOC) and malondialdehyde (MDA) were detected respectively, after vero ceils were treated with different concentrations of ethanol(0, 2.5%, 5%, 10%, 20%) for 3 h, different concentrations of L-carnitine (0, 0.01, 0.1, 1, 10 mmol/L) for 12 h, and different concentrations of L-carnitine pretreated oxidative damaged vero cells for 12 h. The results showed that the activities of SOD and GPx were significantly decreased (P〈0.05), while the levels of MDA were significantly increased (P〈0.05), when vero cells were treated with 5%, 10%, 20% concentration of ethanol. When L-carnitine's concentrations were 1 and 10 mmol/L, the activities of SOD and GPx were significantly increased (P〈0.05). At the same time, the enzyme activities, in vero cells when treated with 1 and 10 mmol/L L-carnitine , of SOD, GPx and the levels of T-AOC were significantly increased (P〈0.05), and the levels of MDA significantly reduced (P〈0.05). In conclusion, 1-10 mmol/L L-carnitine exhibited protective effects on vero cells against ethanol-induced oxidative stress damage. The mechanism might be related to increasing antioxidant capacity of cells and reducing free radicals.
