电针对APP/PS1小鼠行为学及大脑皮层Klotho基因表达的影响 被引量:7

Effects of Electro-Acupuncture on Behavioral Changes, Klotho Level in the Cerebral Cortex in APP/PS1 Transgenic Mice

摘要 本研究通过观察APP/PS1小鼠大脑皮层Klotho基因的表达及其空间学习记忆能力,探讨电针治疗阿尔兹海默病(AD)的机理。方法:APP/PS1双转基因小鼠,随机分为模型组、电针组(百会穴、印堂穴、人中穴),以C57BL/6野生型小鼠作为对照组;采用Morris水迷宫进行空间记忆行为学测试,以免疫组化法观察皮层Klotho蛋白表达趋势,real-time PCR技术检测大脑皮层Klotho m RNA表达水平。结果:与对照组相比,模型组逃避潜伏时间增加、平台穿越次数、平台象限游泳路程减少;与模型组相比,电针组逃避潜伏时间明显减少。与正常组相比,模型组大脑皮层Klotho m RNA表达减少;与模型组相比,电针组Klotho m RNA表达增加。Real time PCR结果与免疫组化所示表达趋势一致。结论:电针可提高APP/PS1小鼠的空间记忆能力,其机制可能与上调Klotho m RNA表达有关。 This aimed to elucidate the mechanism behind the electro-acupuncture (EA)in treating Alzheimer's disease (AD) by observing Klotho mRNA expression in the cerebral cortex and spacial learning and memory in APP/PS1 transgenic mice. APP/PS1 mice were randomly divided into the model group and the EA group, while C57BL/6wild type mice was taken as the control group. Spacial learning and memory was observed by Morris water maze test. Klotho protein expression in the cerebral cortex was detectedy by immunohistochemistry,while its mRNA levels were quantified by real-time PCR. As a result, the escape latency in the model group increased,the number of crossing and the swimming distance in the model group reduced compared with the control group. The escape latency inthe EA group was shorter than that in the model group. Klotho mRNA expression in the cerebral cortex in the model group were lower than that in the control group, while that in theEA group increased. The outcomes of real-time PCR chimed with those of immunohistochemistry. In conclusion,it was demonstrated that EA treatmentprobably improved the spacial learning and momery of the APP/PS1 transgenic mice by upregulatingKlotho expression in the cerebral cortex.
