摘要
介绍了普通烟草中烟碱向降烟碱转化的生物合成过程、遗传机理与NND基因的假基因化研究进展,系统整理和总结了烟草中5个细胞色素P450家族基因的特征特性,并对亚甲基四氢叶酸还原酶基因(Methylenetetrahydrofolate Reductase,MTHFR)间接调控烟碱转化过程进行了深入分析,同时分析了烟碱转化率在不同烟草类型间存在差异的原因及烟草生物碱谱进化的动力,最后结合中国烟草基因组计划对后续研究思路包括新基因挖掘与低烟碱转化烟草分子育种进行了讨论和展望。
Biosynthetic process of genetic mechanisms of nicotine to nornicotine conversion in Nicotiana tabacum L. and research progress of non-functionalization of N-demethylase(NND) genes were introduced in detail in this paper. Genetic features of five identified CYP82E2 family members, namely CYP82E2, CYP82E3, CYP82E4v1, CYP82E5v2 and CYP82E10 belong to cytochrome P450 genes were summarized and in-depth analysis of the function of methylenetetrahydrofolate reductase gene(MTHFR) in indirectly affecting nicotine conversion by regulating CYP82E4v1 negatively was conducted. Reasons for differences of nicotine to nornicotine conversion rate(NCR) in different tobacco species, especially between burley tobacco and flue-cured tobacco, and driving forces for the evolution of alkaloid profile of tobacco was investigated. Prospects of discovering new genes controlling nicotine conversion and developing tobacco varieties with low NCRs by molecular breeding were discussed.
