摘要
为了研究继代次数对暗褐网柄牛肝菌(Phlebopus portentosus)菌种活性的影响,选取生产上优良性状发生退化的栽培菌株11023为研究对象,监测其及继代2次,5次,8次后菌种的各项生理生化特征以及出菇性状。结果显示,随着继代次数增加,菌丝长势减弱,菌落生长速率降低,菌落颜色变浅,菌落边缘整齐度降低;淀粉酶分泌能力减弱;出菇率和单菇重均减小,抗杂能力减弱。推断菌株11023继代次数不宜超过2代。
Mycelial and fruit body growth characteristics of Phlebopus portentosus strains 11023-2,11023-5and 11023-8(derived from cultivar 11023 after two,five and eight sub-cultures for 28-30 days at 30 ℃,respectively on a medium consisting of 200 g potato,20 g glucose,2.0g yeast extract,1.0g MgSO_4·7H_2O,1.0g KH2PO4,15 g agar and 1000 mL ddH_2O)were determined using cultivars 11023 and 13012as controls.Increasing frequency of sub-culture resulted in decreasing mycelial growth rates and vigor,lighter colored mycelia,more irregular-edged fungal colonies,and smaller fungal colony:colored zone diameter ratios in Congo red and Lugol's iodine starch tests,compared with controls.In cultivation tests using artificial logs composed of 10 kg rubber tree(Hevea brasiliensis)sawdust,5kg rice seeds,15.0g MgSO_4,5.0g KH_2PO_4 and 60% water,adjusted to pH 4-6,increasing frequency of sub-culture resulted in lower fruiting rates and single mushroom fruit body weights,and higher contamination rates,compared with controls.
