摘要 采用RT-PCR和Western-blot方法分别检测蝎毒对Hela细胞p21基因的mRNA和蛋白表达,研究蝎毒抗癌作用的分子机理。结果表明:经不同质量浓度(50,100,200μg/m L)蝎毒处理的与未经蝎毒处理的Hela细胞p21基因在mRNA水平表达相近,而在蛋白表达水平上,经蝎毒处理的与未经蝎毒处理的细胞相比,p21基因表达均有明显增强,并以经100μg/m L蝎毒处理的Hela细胞p21基因的蛋白表达增强最为明显,由此证明蝎毒抑制Hela细胞增殖的机理可能是增强了p21基因的蛋白表达。 Hela cells were cultured with scorpion venom and the expression levels of p21 gene was analyzed by RT-PCR and western-blot. The results indicated that the mRNA expression levels of p21 gene in Hela cells dealt with different concentrations of scorpion venom (50, 100, 200 μg/mL) were similar to the ones in normal Hela cells. But the protein expression levels of p21 gene in Hela cells were obviously up-regulated by scorpion venom. And the protein expressions of p21 gene in Hela cells treated with 100 μg/mL scorpion venom were the most up-regulated. These results suggest that the possible mechanism of the scorpion venom is to activate the protein expression of p21 gene.
