核桃叶提取液对CCl_4诱发小鼠急性肝损伤的保护作用 被引量:2

摘要 目的:观察核桃叶提取液对四氯化碳(CCl_4)诱发小鼠急性肝损伤的保护作用。方法:取昆明种小鼠50只,随机均分为正常对照组、模型对照组(给予生理盐水灌胃)及核桃叶低、中、高浓度组(分别给予0.1、0.2、0.4 g/ml的核桃叶提取液灌胃),灌胃量按10 ml/kg体重计算,每天灌胃1次,共14 d。在末次灌胃1 h对正常对照组(给予生理盐水)、模型对照组和核桃叶低、中、高浓度组(给予0.1%CCl_4)腹腔注射,注射量按10 ml/kg的体重计算,24 h后检测小鼠肝匀浆总蛋白(TP)浓度和谷丙转氨酶(GPT)活性、血清GPT活性。结果:肝匀浆TP浓度和GPT活性在各组间两两比较差异均无统计学意义(P>0.05),模型对照组和核桃叶低浓度组的血清GPT活性明显高于正常对照组,差异均有统计学意义(P<0.01或P<0.05);核桃叶中浓度组和高浓度组的血清GPT活性与正常对照组比较差异均无统计学意义(P>0.05);核桃叶低、中、高浓度组和模型对照组的组间两两比较差异均无统计学意义(P>0.05);随着核桃叶提取液浓度的增加血清GPT活性趋向正常对照组。结论:血清GPT活性升高是肝细胞受损的早期敏感指标,核桃叶提取液达到一定浓度具有一定预防自由基破坏肝细胞的功能。 Objective:To observe the protection function of walnut leaf extract solution on the CCl_4 inducing acute liver injury to the mice.Method:50 Kunming mice were randomly divided into normal control group,model control group(normal saline irrigation) and walnut leaves extraction with low,medium and high concentration groups(irrigate the walnut leaf extract solution 0.1 g/ml,0.2 g/ml,0.4 g/ml respectively),and irrigate once a day for 14 days at the amount of irrigation 10 ml/kg which was calculated by the body weight.During the last irrigation which lasted for 1 hour intraperitoneal injection was given to the normal control group(given normal saline),model control group and walnut leaves extract solution with low,medium,high concentration group(given 0.1%CCl_4),the injection quantity was calculated according to 10 ml/kg of the body weight.After 24 hours,the concentration of the total protein(TP) and the activity of the alanine transaminase(GPT),the activity of GPT in serum were detected.Result:The concentration of the TP and the activity of the GPT in liver homogenate were not significantly different between the two groups(P0.05),and the activity of the serum GPT in the model control group and the walnut leaves low concentration group were significantly higher than those in the normal control group,the differences were statistically significant(P0.01 or P0.05).The differences in serum GPT between walnut medium,high concentration group and the nermal control group were not statistically significant(P0.05).The differences among walnut leaves low,medium and high concentration group and model control group were not statistically significant(P0.05).With the increasing of the concentration of the extraction of walnut leaves,the activity of serum GPT became more and more normal.Conclusion:The activity of the serum GPT is a sensitive indicator of the early damage of liver cells,and the extraction of walnut leaves owns certain function to prevent free radical damaging liver cells
