摘要
目的验证大鼠大脑皮质星形胶质细胞中的α2A肾上腺素能受体(α2A-AR)的表达情况,并与α2B肾上腺素能受体(α2B-AR)、α2C肾上腺素能受体(α2C-AR)相比较。方法获取目的基因α2A-AR、α2B-AR、α2C-AR的RNA反转录,与内参基因β-actin一起进行实时荧光定量PCR。重复6组,所得数据进行统计学分析并运用2-△△Ct的方法进行计算。结果大鼠大脑皮质星形胶质细胞中确实表达了α2A-AR,并且α2A-AR的相对表达量是α2B-AR的24.53倍,是α2C-AR的27.98倍。结论α2A-AR可能为大鼠大脑皮质星形胶质细胞接受儿茶酚胺类神经递质调控的主要受体。
Objective To testify the expression of α2A adrenergic receptor(α2A -AR)in the cerebral astroeytes of rats, and compare the expression level of α2A adrenergic receptor with the cc2B adrenergic receptor( α2B - AR) and α2C adrenergie receptor( α2c-AR). Methods Cerebral astroeytes were get from SD rats (aged 1 -3 days). High purity of astrocytes were tested. We extracted and reversely transcribed the target RNA of α2A -AR, cx2B -AR and α2C -AR from the astrocytes, then Real -time PCR was performed to- gether with the house gene β - actin. The aborer proledures were repeated six times, and all data were used for statistical analysis and a method ofio caculate the results. Results α2A - AR were found markablely expressed in the cerebaral astrocytes of rats, which was 24.53 times to the expression of cc2B - AR and 2TM times to the expression of α2C - AR. Conclusion Based on the results, we infer that the α2A - AR is the main adrenergie receptor in the eerebaral astrocytes of rats that receiving eatecholamine regulation.
