摘要 目的探讨不同浓度白藜芦醇(resveratrol,Res)对培养大鼠脑皮层星形胶质细胞增殖及谷氨酸代谢功能的影响。方法原代提取并纯化培养脑皮层星形胶质细胞。采用5-溴脱氧尿核苷(BrdU)掺入法检测细胞增殖;通过同位素掺入法和ELISA法检测谷氨酸代谢指标:谷氨酸摄取量、总谷胱苷肽(GSH)含量和谷氨酰胺合成酶(Glutamine Synthetase,GS)含量与活性。结果Res抑制细胞增殖并随着浓度升高和培养时间延长而抑制增强。Res浓度为25μM和50μM时,谷氨酸摄取量和GSH含量显著增加(P<0.05)。而大于250μM时,两者却降低,但在此高浓度下,GS的细胞含量与活性却显著升高(P<0.05),而在低浓度时无任何变化。结论Res可抑制星形胶质细胞增殖;对其谷氨酸代谢具有二元效应,即低浓度可促进谷氨酸代谢,高浓度则降低谷氨酸代谢。 Objective To investigate the effect of different concentrations of resveratrol on the cell proliferation and the function of glutamate metabolism in primary cortical astrocytes of rats.Methods Astrocyte cultures were prepared of cerebral cortex from neonate SD rats.The proliferation was measured by the incorporation of BrdU;Glutamate uptake was measured us-ing H3-glutamate;the activity of glutamine synthetase and content of glutathione were measured by enzymatic colorimetric assays;the content of glutamine synthetase was measured by ELISA.Results The proliferation was decreased,and followed by the con-centration and the time.At concentration of 25μM and 50μM,resveratrol was able to increase glutamate uptake and glutathione content,but at 250μM,they decreased.But at this high concentration,glutamine synthetase activity was increased.Conclusion The resveratrol can inhibit cell roliferation;and it has dual effect on glutamate uptake,depending on its concentration:low con-centration can promote the glutamate metabolism,and high concentration is opposite.
