摘要
[目的]探讨肠复康方对急性放射性肠炎大鼠一般情况及小肠黏膜相关炎性因子的影响。[方法]以X线直线加速器给予实验大鼠全腹照射(DT9.0Gy),建立急性放射性肠炎大鼠模型。实验大鼠随机分成正常对照组(NC)、模型对照组(MC)、肠复康方低剂量治疗组(CFKD-LG)、肠复康方中剂量治疗组(CFKD-MG)、肠复康方高剂量治疗组(CFKD-HG)。造模成功后连续30d灌胃,期间观察对比大鼠精神状态、摄食进水、排便情况及体质量变化,处死后取相应部位的小肠用RT-PCR方法检测黏膜上皮细胞中炎性因子IL-1β、IL-6、TNF-αmRNA的表达。[结果]造模成功后肠复康方低、中、高剂量组精神状态、饮食进水、排便情况较模型对照组好;与模型对照组比较,肠复康方低、中、高剂量组大鼠体质量显著增加,肠复康方中剂量组效优(P<0.01)。肠复康方低、中、高剂量组小肠黏膜的炎性因子IL-1β、IL-6、TNF-αmRNA较模型对照组显著下调(P<0.01)。[结论]肠复康方低、中、高剂量组通过下调小肠黏膜上皮细胞中炎性因子IL-1β、IL-6、TNF-αmRNA的表达,改善小肠黏膜的炎症反应,促进急性放射性肠炎受损小肠黏膜的修复,以中剂量效优。
[Objective]To explore the effect of Changfukang decoction on general condition and the expression of inflammatory factor of small intestinal mucosa in rats with acute radiation enteritis.[Methods]An abdominal irradiation in a dose of 9.0Gy X-ray of linear accelerator was performed once on a group of rats to establish a model of acute radiation enteritis.The experimental rats were randomly divided into five groups:normal control group,model control group,Changfukang decoction low dose group,middle dose group and high dose group.After the model was established,all rats were given corresponding treatment by gavage once a day for 30 days.In the meantime,the mental state,dietary intake,defecation and body mass changes of all rats were observed and compared.After the rats were sacrificed,corresponding intestinal tissues were taken for detecting the expression of inflammatory related factors IL-1β,IL-6and TNF-αmRNA in the intestinal mucosal epithelial cells by RT-PCR.[Results]After the model was established,medicationgroups' mental state,dietary intake and defecation were better than model control group.Compared with model control group,medication groups had significant increase in body mass,typically in Changfukang decoction middle dose group(P〈0.01).Treatment with low,middle and high dose of Changfukang decoction also significantly down-regulated the expression of IL-1β,IL-6and TNF-αmRNA in the intestinal mucosal epithelial cells compared with that of model control group(P〈0.01).[Conclusion]The low,middle and high dose group of Changfukang decoction improved the small intestinal mucosa inflammation and promoted restoration of the acute radiation enteritis of small intestinal mucosa damage by down-regulating the expression of IL-1β,IL-6and TNF-αmRNA in the intestinal mucosal epithelial cells.The effect is especially better in middle dose group.
