摘要
目的观察有丝分裂前期检查点基因CHFR在食管癌、食管上皮内瘤变及正常食管组织中蛋白的表达,了解食管癌中CHFR蛋白表达与患者临床病理资料之间的关系,探讨其在食管癌发生发展过程中的作用及相关的分子机制。方法收集食管癌标本92例以及相配对的食管上皮内瘤变11例,另取正常食管组织27例。采用免疫组化检测92例食管癌及其对应的食管上皮内瘤变、正常食管组织中CHFR蛋白的表达。通过免疫组化半定量积分法判断免疫组化结果。结果食管癌组织中CHFR蛋白阳性表达率为7.61%(7/92),低于正常食管组织的96.3%(26/27)和食管上皮内瘤变组织的54.55%(6/11),且差异有统计学意义(P﹤0.05);食管上皮内瘤变组织中CHFR蛋白阳性表达率也低于正常食管组织,且差异有统计学意义(P﹤0.05)。高分化的食管癌组织中CHFR蛋白表达阳性率的33.33%(6/18)明显高于中-低分化的食管癌组织中CHFR蛋白表达阳性率的1.35%(1/74),差异具有统计学意义(P﹤0.05)。患者的不同年龄、性别以及其他病理资料之间CHFR蛋白表达阳性率比较,差异无统计学意义(P﹥0.05)。结论食管癌中有丝分裂检查点基因CHFR蛋白的表达下调或缺失是频发、早期事件,可能参与食管癌的发生发展,其与食管癌分化程度的关系可能更为密切。
Objective To observe the protein expression of CHFR prophase check point gene in esophageal cancer, esophageal intraepithelial neoplasia, and normal esophageal tissues, discuss the relationship between the protein expres-sion of CHFR in esophageal cancer and clinicopathologic characteristics, and to explore the correlating molecular mecha-nism of CHFR expression in esophageal carcinogenesis and development. Method 92 specimens of esophageal cancer, 11 matched samples of esophageal intraepithelial neoplasia, and 27 samples of normal esophageal tissues were collected, in which the protein expression of CHFR were detected immunohistochemically. Semi-quantitative immunohistochemical analysis was applied to evaluate the immunohistochemical results. Result The positive rate of CHFR protein expression was 7.61%(7/92) in esophageal cancers, and was significantly lower than that in normal esophageal tissues at 96.3%(26/27) and in esophageal intraepithelial neoplasia at 54.55%(6/11) (P〈0.05), respectively;And the positive expression of CHFR in esophageal intraepithelial neoplasia was also lower as compared with that in normal tissues (P〈0.05);In well differentiat-ed esophageal cancer tissues, the positive rate of CHFR protein expression was 33.33%(6/18), and was significantly high-er than that in moderately or poorly differentiated esophageal cancers, with significant difference observed (P〈0.05);No obvious correlation was observed between the CHFR protein expression and age, gender as well as other pathologic char-acteristics (P〉0.05). Conclusion Down-regulation or absence of mitotic checkpoint CHFR protein expression is a fre-quent and early event, which may be involved in the esophageal carcinogenesis and is potentially yet closely related with the differentiation of esophageal cancers.
