摘要
目的探讨膜联蛋白Ⅱ(AnnexinⅡ)在胃癌组织中的表达及其在胃癌转移中的作用。方法采用免疫组织化学法检测AnnexinⅡ在51例胃癌组织和24例癌旁组织中的表达,并统计AnnexinⅡ阳性表达与胃癌患者临床特征的关系。用AnnexinⅡ特异性si RNA在胃癌HGC-27细胞抑制AnnexinⅡ的表达,应用黏附实验、侵袭实验检测抑制AnnexinⅡ表达后对胃癌细胞的黏附、侵袭能力的影响。结果 AnnexinⅡ蛋白在胃癌组织中阳性率为82.4%,在癌旁组织中阳性率为37.5%,两组比较差异有统计学意义(P﹤0.01);并且AnnexinⅡ阳性表达与患者的性别与年龄无关(P﹥0.05),而与肿瘤大小、组织分化程度、临床分期、淋巴结转移临床特征有关,差异具有统计学意义(P﹤0.05)。在胃癌HGC-27细胞转染AnnexinⅡsi RNA后,细胞中AnnexinⅡ的蛋白和m RNA水平均下降,差异有统计学意义(P﹤0.05)。并且抑制AnnexinⅡ表达后,胃癌HGC-27细胞的黏附、侵袭能力下降,差异有统计学意义(P﹤0.05)。结论 AnnexinⅡ在胃癌组织中高表达,AnnexinⅡ蛋白表达与肿瘤大小、组织分化程度、临床分期、淋巴结转移临床特征有关,并且AnnexinⅡ高表达可促进胃癌转移。因此,AnnexinⅡ可以作为抑制胃癌转移的潜在靶点,开发靶向AnnexinⅡ的药物可能成为治疗胃癌转移的新方法。
Objective To investigate the significance and the role of Annexin II expression in gastric cancer tissues and gastric cancer metastasis. Method Expression of annexin II in 51 cases of gastric cancer and 24 cases of adjacent normal gastric tissues was detected by immunohistochemistry staining, then the correlation between positive expression of Annexin II and clinical characteristics of gastric cancer patients were summarized and analyzed. Annexin II specific siRNA interference was established in human gastric cancer HGC-27 cells to downregulate the expressions of Annexin II, and cell adherence test and cell invasion test were performed to evaluate the adhesion and invasion potential of gastric cancer cells as Annexin II expression was inhibited. Result The positive expression rate of Annexin II in gastric cancer tissues was 82.4%, significantly higher than that in the adjacent normal gastric tissues at 37.5%, with significant differ-ence observed (P〈0.01);Besides, the positive expression of Annexin II was independent of gender or age of gastric can-cer patients (P〉0.05), while was correlated with tumor size, histological differentiation, clinical TNM stage, and lymph nodes metastasis (all P〈0.05). The transfection of Annexin II siRNA in human gastric cancer HGC-27 cells led to signifi-cant decreases of Annexin II protein and mRNA (P〈0.05), and as the Annexin II expression was downregulated, the adhe-sion and invasion potential of human HGC-27 gastric cancer cells were significantly decreased (P〈0.05). Conclusion Annexin II is over-expressed in gastric cancer tissues and the positive expression of Annexin II is correlated with tumor size, histological differentiation and lymph node metastasis, and the high expression of Annexin II may promote the me-tastasis of gastric cancer, so Annexin II may be an alternative and potential target for preventing metastasis of gastric can-cer, and annexin II targeted medication is a promising option in countering cancer metastasis.
