摘要
目的比较牛膝醇提物(AEAB)与硫酸氨基葡萄糖和双醋瑞因对于兔骨关节炎(OA)的疗效。方法将35只实验兔采用随机区组分组法分为正常组、模型组、AEAB组、硫酸氨基葡萄糖组和双醋瑞因组,每组7只。石膏固定制动6周制备OA模型,造模后,分别以蒸馏水、AEAB、硫酸氨基葡萄糖及双醋瑞因灌胃,正常组不予处理,6周后处死实验兔,取软骨标本进行大体观察、Pelletier评分、Mankin评分,用免疫组化测定软骨Ⅱ型胶原蛋白含量,用酶联免疫吸附测定(ELISA)法检测血清和关节液中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)以及基质金属蛋白酶-3(MMP-3)的含量。结果 AEAB组、硫酸氨基葡萄糖组和双醋瑞因组Pelletier评分、Mankin评分、Ⅱ型胶原蛋白含量、血清和关节液中TNF-α、IL-1β以及MMP-3的含量分别与正常组、模型组比较,差异均有统计学意义(P<0.05或P<0.01);AEAB组与硫酸氨基葡萄糖组及双醋瑞因组比较,上述指标差异均无统计学意义(均P>0.05)。结论 AEAB可以促进兔OA模型的软骨修复,其疗效与硫酸氨基葡萄糖及双醋瑞因相当,具有较高的临床应用价值。
Objective To compare the effects of alcohol extract of A chyranthis Bidentatae Radix （AEAB）, glucosamine sulfate and Diacerein in the treatment of rabbits with osteoarthritis （OA）. Methods With blocked randomization, thirtyfive experimental rabbits were divided into normal group, model group, AEAB group, glueosamine sulfate group, Diacerein group, with 7 rabbits in each group. The OA models were established by immobilizing rabbits, knees with plaster cast for 6 weeks, and distilled water, AEAB, glucosamine sulfate, Diacerein were respectively given by lavage after modeling. Normal group was not treated. The experimental rabbits were killed six weeks later, and the cartilage samples were taken to perform gross observation, Pelletier score, Mankin score, and the type Ⅱ collagen was measured by im- munohistochemistry method and the tumor necrosis factor alpha （TNF-α）, interleukin-1β （IL-1β） and matrix metallo- proteinase-3 （MMP-3） in serum and joint fluid were detected by enzyme-linked immunosorbent assay （ELISA） method. Results There were all statistically significant differences of Pelletier score, Mankin score, content ＇of type II collagen, TNF-ot, IL-1β and MMP-3 in AEAB group, glucosamine sulfate group, Diacerein group compared with normal group and model group respectively （P 〈 0.05 or P 〈 0.01）, while there were no statistically significant differences in afore- mentioned indexes among AEAB group, glucosamine sulfate group and Diacerein group respectively （all P 〉 0.05）. Conclusion AEAB can promote the cartilage repair in rabbits with OA, and it has equivalent effects with glucosamine sulfate E/nd Diacerein, so it has high value in clinical application.
