摘要
目的:研究温经消痤颗粒对变应性接触性皮炎(ACD)小鼠模型的抗炎效果和免疫机制。方法:60只ICR小鼠随机分成空白组、模型组、丹参酮(0.8g/kg)组及温经消痤颗粒低、中、高剂量(10.3、20.6、41.2g/kg)组,每组10只。除空白组外其余各组以2,4-二硝基氟苯(DNFB)建立小鼠ACD模型,空白组及模型组予蒸馏水灌胃,其余各组分别以相应药物灌胃。实验结束后,观察小鼠耳组织病理、耳肿胀度、肿胀抑制率、脏器指数及血清中IL-2β、TNF-α、IL-10的含量。结果:与空白组比较,各组耳肿胀度、脾脏指数、IL-2β、TNF-α均显著升高(P<0.05,P<0.01);与模型组比较,各给药组耳肿胀度、脾脏指数、IL-2β、TNF-α均显著降低(P<0.05,P<0.01),耳组织水肿及炎性细胞浸润均减轻;与丹参酮组比较,高剂量组耳肿胀度、脾脏指数、IL-2β均显著降低(P<0.01)。结论:温经消痤颗粒具有良好的抗炎作用,其抗炎机制可能与抑制免疫反应有关。
Objective:To research effects of Wenjing Xiaocuo Particle on inhibiting inflammation of allergic contact dermatitis(ACD) mouse model and explore its mechanism.Methods:Sixty ICR mice were randomly divided into:control group,model group,tanshinone group,Wenjing Xiaocuo Particle low,medium and high dose(10.3,20.6,41.2g/kg) group,10 rats per each group.DNFB was used to establish an ACD mice model.The control group and model group were given distilled water by gavage.The others were given the corresponding drugs by gavage.After experiment,ear tissues were removed to calculate ear swelling degree,swelling inhibition rate,pathological changes,and spleen and thymus were removed to determine the organ index.The level of IL-2β,IL-10 and TNF-α in serum were detected by ELISA assay.Results:Compared with control group,the ear swelling degree,the spleen index,the levels of IL-2β and TNF-α were significantly increased in other groups(P〈0.05,P〈0.01).Compared with model group,the ear swelling degree,the spleen index,the levels of IL-2β and TNF-α were significantly decreased in treatment groups(P〈0.05,P〈0.01),meanwhile,ear tissue edema and inflammatory cells ini ltration were reduced.Compared with tanshinone group,the ear swelling degree,the spleen index and the levels of IL-2β were significantly decreased in Wenjing Xiaocuo Particle high dose group(P〈0.01).Conclusion:Wengjing Xiaocuo Particle had a good anti-inflammatory effects,its anti-inflammatory mechanism may related to inhibiting the immune response.
