摘要
对生物酶法转化西洋参多糖的产物进行分离纯化得到寡糖,并对其进行鉴定。西洋参多糖的酶解产物经Sevage法除蛋白、Sephadex G-75葡聚糖凝胶柱层析分离纯化,得到2个组分PP-1、PP-2相对分子质量约为7.5×10~4和6.7×10~3,其中PP-2为酶降解寡糖。HPLC法对PP-2进行鉴定,结果表明,PP-2为均一的寡糖,其单糖组成为葡萄糖。高碘酸氧化、Smith降解、红外光谱对PP-2的结构进行鉴定,表明PP-2为主链由α-D-(1→4)和α-D-(1→6)吡喃葡萄糖构成的葡聚糖。
Oligosaccharide was obtained by enzymatically degrading polysaccharides in Panax quinquefolium. After removed proteins components by Sevage method, two fractions PP-1 and PP-2 were obtained by Sephadex G-75, relative molecular weights of which were about 7.5 × 104 and 6.7 × 103. PP-2 was a homogeneous oligosaccharide from enzymatic hydrolysis, which was mainly composed of glucose. Results of Malaprade reaction, Smith degradation and IR suggested that PP-2 was glucan and mainly composed of α-D-(1 → 4) and α-D-(1 → 6) pyran glucose.
