响应面法优化西洋参多糖的酶解辅助提取工艺 被引量:14

Optimization of Enzymatic Extraction of Polysaccharides from Panax quinquefolium by Response Surface Methodology

摘要 为研究西洋参中多糖的提取工艺条件,以西洋参为原料,利用纤维素酶和木瓜蛋白酶对其进行酶解,采用酶解辅助提取西洋参多糖,以酶解温度、酶解时间、pH、复合酶添加量、复合酶配比等对酶解辅助提取法进行单因素试验,初步确定各因素的最佳范围。在单因素试验基础上,采用响应曲面试验设计确定酶解辅助提取最佳条件。经试验得到,最优工艺参数为:酶解时间1.37 h、酶解温度60.20℃、pH 6.50、酶添加量1.44%,此条件下多糖得率为13.77%。酶解辅助提取方法简单、效率高,可作为西洋参多糖的提取工艺。 Study the condition of extraction process of polysaccharide from Panax quinquefolium.Taking Panax quinquefolium as the raw material,use cellulase and papain to hydrolysis them.The enzymatic assisted extraction of polysaccharide from Panax quinquefolium.By making the single factor experiments,the optimum technological parameters for enzymatic assisted extraction were obtained.On the basis of the single factor experiment,response surface methodology was used to optimize process condition.The results showed that the optimum extraction condition was found to be 1.37 h,60.20℃,pH 6.50 and an enzyme dosage of 1.44%.Under this condition,the polysaccharide yield was 27.54%.This extraction technique was simple,efficient and applicable for the extraction of polysaccharides from Panax quinquefolium.
