摘要
以农作物副产品玉米秸秆为研究对象,采用水解纤维素性能优良的绿色木霉(13001)及高产蛋白质的产朊假丝酵母(1769)为发酵生产菌株共同处理,生产蛋白质含量较高,富含多种矿物元素的单细胞蛋白产品。探讨了玉米秸秆预处理方法、绿色木霉接种工艺、纤维素降解工艺、不同氮源对玉米秸秆发酵糖化的影响,以及单细胞蛋白发酵工艺,通过对比试验确定了玉米秸秆最佳的预处理方法为粉碎后加入氨水对玉米秸秆进行预处理、接种6%绿色木霉(13001)产酶效率最高、添加15%纤维素酶曲水解24h纤维素水解最完全、加入氯化铵作为氮源发酵糖化效果最好,通过正交试验确定了单细胞蛋白发酵的最适工艺条件为加入3倍水,温度控制在29~31℃,起始pH控制在5.5左右,发酵时间48h左右。
Using maize straw as main research objects, the hydrolysis of cellulose with excellent performance of Trichoderma viride(13001) and high-yield protein of Candida utilis yeast strains (1769) for fermentation production processing together, producting the single cell protein products of high protein content and rich in various mineral elements. Discussed the pretreatment methods of maize stalk, the inoculation technology of green wood mold, the cellulose degradation process, the effects of different nitrogen sources on the fermentation of maize straw and single cell protein fermentation process, by contrast experiments, the best pretreatment method of maize stalk was pretreated by adding ammonia, the highest efficiency of enzyme production was adding 6% trichoderma viride(13001), the hydrolysis of 24 h with the addition of 15~ cellulase was the most complete, the best effect of the fermentation was added ammonium chloride as the nitrogen source were determined, by orthogonal test to determine the optimum fermentation technological conditions of single cell protein is added in 3 times of water, temperature controlled in 29~31 *C, starting pH at about 5. 5, fermentation for 48 hours.
