摘要
目的:观察青葙子提取物(CAE)对链脲佐菌素(STZ)诱导的小鼠糖尿病视网膜病的改善作用,并研究其可能的机理。方法采用STZ诱导C57小鼠发生糖尿病(DM),进而诱导产生糖尿病视网膜病(DR),然后给予DM小鼠CAE干预。通过视网膜组织HE染色来检测视网膜血管新生情况;采用ELISA和Western-blot分别检测玻璃体和血清中VEGF水平以及视网膜组织HIF-1α-VEGFA通路表达情况。结果:CAE(30、100 mg·kg-1)能够明显降低DM小鼠视网膜血管新生情况;研究其机理发现CAE能够下调DR小鼠升高的血清中VEGFA水平,CAE能够抑制DR小鼠视网膜组织HIF-1α入核作用并下调DR小鼠视网膜组织中升高的VEGF表达。结论:CAE具有改善DR的药效活性,其机理为CAE通过抑制DR小鼠视网膜组织HIF-1α核转录作用,进而降低DR小鼠血清和视网膜组织中VEGFA表达。
Objective: To investigate the amelioration of Celosia argentea L. extract (CAE) on streptozotocin (STZ)-induced DR and its engaged mechanism. Methods: STZ-induced diabetic retinopathy by injection in C57BL/6 and then the diabetic mice were given with CAE. Retinal neovascularization was detected by histologi- cal assessment of retinas by H&E staining. The content of VEGF in vitreous and serum was detected by ELISA. The retinal VEGFA expressions and nuclear translocation of HIF-1α were detected by Western-blot analysis. Results: CAE reduced the increased retinal vessels in STZ-induced DR mice. CAE reduced the increased serum VEGFA levels arid retinal VEGFA expressions and inhibited the retinal nuclear translocation of HIF-1α in STZ-induced DR mice. Conclusion: CAE ameliorate DR via inhibiting retinal angiogenesis and reducing the ex- pression of pro-angiogenic cytokine VEGFA.
