摘要
目的观察电针对WKY抑郁模型大鼠行为学及缰核c-fos蛋白表达的影响。方法将24只雄性WKY大鼠随机分为电针组(EA)、假针组(sham EA)、模型组(model),每组8只。另将Wistar雄性大鼠8只设为正常组(normal)。通过旷场实验、糖水实验、强迫游泳实验对大鼠进行行为学评价,并用免疫荧光方法检测大鼠缰核中c-fos表达。结果与正常组比较,模型组旷场实验的水平运动、垂直运动和蔗糖水摄入比均明显下降(P<0.05),强迫游泳的不动时间和缰核c-fos表达均增加(P<0.05)。与假针组比较,电针组旷场实验的水平运动和垂直运动、蔗糖水摄入比均明显增加(P<0.05),强迫游泳的不动时间和缰核c-fos表达减少(P<0.05)。结论电针可改善WKY抑郁模型大鼠的行为异常,明显降低缰核中c-fos的表达,这可能是电针治疗抑郁症的机制之一。
Objective To observe the effects of electroacupuncture (EA) on depressive-like behavior and c-fos protein expression of habenular nucleus in the WKY rat model. Methods 24 male WKY rats were randomly divided into the EA, sham and model groups, 8 in each group. 8 Wistar male rats were set as normal group. The behaviors were evaluated by open field test, sucrose preference test and force swimming test, the expression of c-fos was detected by immunofluorescence. Results Compared with the control group , the behaviors of open field test and Sucrose preference test in model group decreased significantly ( P 〈 0.05 ), and the immobile time of force swimming test and c-los expression of habenular nucleus increased significantly ( P 〈 0.05 ). Compared with sham group , the behaviors of open field test and Sucrose preference test in EA group increased significantly ( P 〈 0.05 ), and the immobile time of force swimming test and c-fns expression of habenular nucleus decreased significantly ( P 〈 0.05 ). Conclusion Electroacupuncture cauld improve the depressive-like behaviors of WKY rats and decrease the expression of c-fos protein in habenular nucleus. It may be one of the mechanisms of electroacupuncture on depression.
