启膈散对食管癌小鼠生存质量及免疫调节的影响 被引量:12

Qigesan impact on quality of life and immune regulation in mice of esophageal cancer

摘要 目的观察启膈散对4NQO诱导食管癌小鼠生存质量、体重、脏器重量和指数、食管组织病理形态、细胞因子及淋巴细胞亚群的干预作用。方法 0.1mg·ml^(-1)4NQO水诱导小鼠食管癌模型,16周后撤除干预因素,观察进食及饮水并称重,32周后处死称重脏器并计算脏器指数,观察食管组织,检测外周血中IL-10、TNF、IFN-γ、MCP-1、IL-6、IL-12p70等细胞因子,检测外周血中淋巴细胞亚群(CD3^+/CD19^+,CD4^+/CD8^+)。结果启膈散组小鼠形体、活动度正常,体重接近正常组;脾、肝重量高于模型组(P<0.05),肾、心、肺、肝指数降低(P<0.05);镜下观食管基底细胞层有轻度增生,角质层液化基本消失,其它均正常;与模型组比可上调IL-10细胞因子(P<0.05),降低CD3^+、CD19^+、CD3^+/CD19^+、CD8^+,提高CD4^+/CD8^+(P<0.05)。结论启膈散能改善生存质量、保护脏器、保护组织病理损害;通过提高IL-10细胞因子水平提升免疫功能,提高CD3^+、CD19^+、CD8^+淋巴细胞,纠正CD4^+/CD8^+比例降低而改善免疫功能失衡及免疫抑制状态。 Objective viscera weight and viscera index,esophageal pathological tissue, plasma cytokines and lymphocyte sub- group of mice of esophageal cancer induced by 4NQO. Methods To make mice drink the water mixed with 0.1 mg · mL-1 4NQO to induct esophageal cancer. All mice were divided into normal group ,model group and Qigesan group , after 16 weeks then remove intervention factors, to observe the diet and weigh mice ;After 32 weeks kill all mice to weigh viscera and calculate viscera index , to observe esophageal pathological tissue, to detect IL - 10,TNF, IFN -γ, MCP - 1 ,IL -6, IL - 12p70 plasma cytokines and lym- phocyte subgroup( CD3 +/CD19 + ,CD3 +/CD4 +, CD3 +/CD8 + ). Results Mice of Qigesan group were normal activity, not obvi- ous declining of weights ; weighs of the spleen, liver were higher than the model group, viscera index of kidney, heart, lung and liver were lower, esophageal tissue of mice were similar to the normal group, esophageal mucosa epithelial basal cell layer represented slight abnormal hyperplasia and others were normal . IL- 10,IFN- γ/plasma cytokines could be adjusted upwards, CD3+, CD19+ , CD3 +/CD19 + , CD8 + , CD4 +/CD8 + could be increased. Conclusion Qigesan can improve the quality of life of the e- sophageal cancer mice,protect viscera and the esophageal pathological tissue, improve the immune function by promoting 1L- 10 and other plasma eytokines and increase CD3 + ,CD19 + ,CD8 + ,put CIM +/CD8 + ratio right .
