摘要
目的:观察石斛合剂含药血清对H_2O_2诱导SH-SY5Y细胞氧化应激的影响及可能的机制。方法:体外培养的SH-SY5Y细胞预先予以石斛合剂含药血清(10%)孵育24 h,加入终浓度为400μmol·L-1的H_2O_2培养1 h,采用MTT法测细胞存活率、酶法测细胞内SOD活力、MDA和GSH含量、流式细胞术测细胞凋亡情况等,以观察石斛合剂含药血清对H_2O_2诱导SH-SY5Y细胞氧化应激损伤的影响。结果:与模型组比较,石斛合剂含药血清能明显提高受H_2O_2刺激的SH-SY5Y细胞的存活率(P<0.05),降低MDA水平和提高SOD和GSH活力(P<0.01),并减少因H_2O_2引起的神经细胞凋亡。结论:石斛合剂可保护H_2O_2引起的SH-SY5Y细胞氧化应激损伤并抑制其凋亡。
Objective: To observe the effect of Dendrobium Compound( DC) on oxidative stress in SH-SY5 Y cells induced by H2O2 and study its possible mechanism. Methods: Hatch SH-SY5 Y cells in vitro with DC-containing serum( 10%) in advance for 24 h,and add H2O2( 400 μmol/L) to culture for 1 h,and then use MTT assay to measure the cell survival rate and enzyme method to measure intra-cellular SOD activity,MDA and GSH content and flow cytometry to test cell apoptosis situation etc.,in order to observe the effect of DC-containing serum on SH-SY5 Y cells oxidative stress injury. Results: Compared to the model group,DC-containing serum could significantly improve the survival rate of SH-SY5 Y cells stimulated by H2O2( P 〈 0. 05),decrease the level of MDA and increase the activity of SOD and GSH( P 〈 0. 01) and reduce the nerve cell apoptosis induced by H2O2. Conclusion: DC can protect SH-SY5 Y cells from oxidative stress damage induced by H2O2 and inhibit cells’ apoptosis.
