摘要
目的探讨硫酸镁(MgSO_4)对大鼠脑缺血后氧化应激和NF-кB蛋白表达的影响。方法 60只SD大鼠随机分为假手术组、模型组及MgSO_430 mg/kg、60 mg/kg和120 mg/kg组。采用线栓法制备大鼠永久性大脑中动脉缺血模型(MCAO)。术后即刻假手术组与模型组腹腔注射生理盐水1 ml,MgSO_4各剂量组按上述剂量腹腔注射250 g/L MgSO_4注射液(均用生理盐水稀释至1 ml)。24 h后检测脑匀浆液中超氧化物岐化酶活性(SOD)、丙二醛(MDA)含量,应用免疫组化技术观察NF-кB蛋白阳性细胞的表达。结果与模型组相比,MgSO_460 mg/kg和MgSO_4120 mg/kg两组的SOD活性均显著增高(P=0.000),而MgSO_430 mg/kg组没有统计学意义;MgSO_4各剂量组的MDA含量均显著降低(P=0.000);MgSO_460 mg/kg组和MgSO_4120 mg/kg组的NF-кB蛋白阳性细胞表达明显减少(P=0.000),而MgSO_430 mg/kg组无统计学差异。与MgSO_430 mg/kg组比较,MgSO_4120 mg/kg组的SOD活性显著增高(P=0.015),而MgSO_460 mg/kg组没有统计学意义;MgSO_460 mg/kg组和MgSO_4120 mg/kg组的MDA含量显著降低(P=0.001和P=0.000);MgSO_460 mg/kg组和MgSO_4120 mg/kg两组的NF-кB蛋白阳性细胞表达明显减少,差异具有显著性(P=0.000)。结论 MgSO_4对大鼠局灶性脑缺血后氧化应激损伤具有良好的保护作用,其机制可能与清除自由基、减轻氧化性损伤和抑制NF-кB蛋白的表达有关。
Objective To investigate the effects of different doses of magnesium sulfate(MgSO_4) on oxidative stress and expression of NF-kappa after focal cerebral ischemia in rats.Methods Sixty SD rats were randomly divided into sham operation group,model group and Magnesium sulfatein group 30,60 and 120 mg/kg treatment groups.Using thread embolism method prepared rat permanent middle cerebral artery ischemia model(MCAO).the sham operation group and model group were given intraperitoneal injection of 1 ml normal saline postoperative immediately.By intraperitoneal injection of MgSO_4 each dose group according to the dose of 250 g/L MgSO_4injection(were diluted with saline to 1ml).After 24 hours,the activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA) were measured in brain homogenate,meanwhile the histopathological changes of cerebral tissues were observed and at the same time the expression NF-kappa B protein positive cells were evaluated in each group by immunohistochemistry.Results Compared with the model group,the activity of SOD in magnesium sulfate 60 and 120 mg/kg groups were significantly higher than those in the model group(P = 0.000),while magnesium sulfate 30 mg/kg had not statistical significance;the MDA content of magnesium sulfate in each dose group were significantly lower than those of the model group(P = 0.000);Magnesium sulfate in magnesium sulfate 60 and 120 mg/kg groups of NF-kappa B protein positive cells decreased significantly(P = 0.000),while magnesium sulfate 30 mg/kg had not statistical significance.Compared with magnesium sulfate 30 mg/kg,the activity of SOD in magnesium sulfate 120 mg/kg increased significantly(P = 0.015),while magnesium sulfate 60 mg/kg had not statistical significance;the MDA content in magnesium sulfate 60 and 120 mg/kg groups were significantly lower than those in the model group(P = 0.001 and P = 0.000),the expression of NF-kappa B protein positive cells in magnesium sulfate 60 and 120 mg/kg groups decreased si
