APOPTOSIS IN RATS WITH MIDDLE CEREBRAL ARTERY OCCLUSION AND ITS MECHANISM

摘要 目的 :研究急性脑缺血边缘系统bcl- 2家族及白介素 - 1β转化酶 (ICE)基因的同期转录变化特征及其与细胞凋亡的关系 ,并探讨牛磺酸对其影响和可能机制。方法 :制做MCAO模型 ,在立体定向仪引导下行侧脑室给药 ,监测空白对照组、生理盐水干预组和牛磺酸干预组的脑梗塞容积、颞皮质、海马和下丘脑的凋亡细胞数、bcl- 2基因阳性转录细胞数和ICE基因阳性转录细胞数的动态变化。结果 :正常脑组织颞皮质、海马和下丘脑 3部位的bcl- 2基因均有密集转录。空白对照组和生理盐水干预组各部位于再灌注 2 4h ,bcl- 2基因转录开始下降 ,急剧下降期位于MCAO后 2 4h~ 4 8h。牛磺酸干预组于再灌注 2 4h开始下降 ,并于以后各时相继续缓慢下降而始终未达低谷。牛磺酸干预组各时相的bcl- 2基因阳性转录细胞数显著高于其它两组 (P <0 .0 5 )。ICE基因的同期转录水平几乎与bcl- 2基因转录水平呈反向变化。结论 :脑缺血及再灌注时bcl- 2基因转录受抑制 ,而白介素 - 1β转化酶基因转录上调 ,牛磺酸有显著减轻这些基因转录、细胞凋亡和缩小脑梗塞容积的作用。 Objective:To investigate the features of transcription of bcl-2 clan and interleukin-1β converting enzyme (ICE) gene in limbic system after rats middle cerebral artery occlusion (MCAO), and effects of taurine on these changes. Methods:We made MCAO models,under the guidance of stereoscopic direction set injected medicine into ventricles,and determined the dynamic changes of cerebral infarction volumes, positive transcription cell counts of apoptosis, bcl-2 clan and ICE gene in temple cortex, hippocampus and hypothalamus during rats MCAO or reperfusion. Results:Bcl-2 gene had concentrated positive transcription in temple cortex, hippocampus and hypothalamus of normal rats. In blank control group and physiological salt solution control group, bcl-2 gene transcription began to decrease at reperfusion 24 hours. Sharp drops period located during reperfusion 24 to 48 hours, after MCAO. In taurine intervene group, at reperfusion 24 hours, bcl-2 gene transcription began to decrease, and slowly drop that not reached the lowest level all along;bcl-2 gene positive transcription cell counts at every times in taurine intervene group were significantly higher than other two groups ( P <0.05). ICE gene transcription level were almost oppositely changed compared with those of bcl-2 gene. Conclusions:During cerebral ischemia and reperfusion, bcl-2 gene transcription is restrained, while ICE gene transcription is up-regulated. Taurine can markedly alleviate these genes transcription and apoptosis, and can shrink cerebral infarction volume.
