补肾活血颗粒对去势骨质疏松大鼠OPG/RANK/RANKL系统的影响 被引量:9

Effects of Bushen Huoxue Granule(补肾活血颗粒) on OPG/RANK/RANKL System in Ovariectomized Rats with Osteoporosis

摘要 目的:观察补肾活血颗粒对去势骨质疏松大鼠骨组织骨保护素(OPG)、核因子κB受体活化因子配体(RANKL)、核因子κB受体活化因子(RANK)mRNA表达及其骨密度的影响,探讨补肾活血颗粒治疗骨质疏松的作用机制,为临床使用补肾活血中药防治骨质疏松症(OP)提供实验依据。方法:将雌性SD大鼠80只随机分为模型组、补肾活血颗粒治疗组、倍美力阳性对照组、正常对照组4组;除正常组外,其余各组大鼠行双侧卵巢去除术,正常组大鼠行假手术处理;各组大鼠给予相应处理12 W。采用双能X线骨密度仪测定大鼠右股骨上干骺端骨密度、HE染色观察骨组织形态改变、免疫组织化学染色方法及荧光定量PCR检测骨组织OPG、RANK、RANKL相关蛋白和基因的表达。结果:补肾活血颗粒能够显著增加去势骨质疏松大鼠的骨密度;提高其骨组织OPG、RANK mRNA及蛋白的表达水平,降低RANKL mRNA及蛋白的表达水平;作用与阳性对照组相当。结论:补肾活血颗粒可显著增强去势骨质疏松大鼠的骨密度,促进OPG、RANK mRNA和蛋白的表达及抑制RANKL mRNA和蛋白表达是其部分作用机制。 Objective: To observe the effects of Bushen Huoxue Granules on OPGmRNA,RANKLmRNA and RANKmRNA expressions and the bone mineral density( BMD) in ovariectomized rats,to explore the mechanism of Bushen Huoxue Granules on the treatment of osteoporosis. Methods: 80 female rats were randomly divided into four groups,model group,Bushen Huoxue Granules group,Premarin group( positive group) and normal group,all rats were ovarietomized except normal group with sham operation.,each group was given the corresponding medicine or distilled water for 12 weeks. BMD of the right femoral metaphysis was determined by DEX- L,the morphological changes of bone tissue were observed by microscope,expression levels of Canonical OPG / RANK / RANKL system proteins and genes were analyzed by immumohistochemical staining and RT- PCR respectively. Results: Bushen Huoxue Granules could enhance BMD in ovariectomized rats with osteoporosis,at the same time it also rise the expressions of OPG mRNA,RANK mRNA and their protein,reduce the expressions of RANKL mRNA and protein.,the effects of Bushen Huoxue Granule were semilar to the positive medicine. Conclusion: Bushen Huoxue Granules has obvious therapeutic effect on osteoporosis in ovariectomized rats,promoting the expressions of OPG mRNA,RANK mRNA and inhibiting the expression of RANKL mRNA were the part of mechanism.
