摘要
目的:观察艾灸预处理对大鼠脑缺血再灌注损伤的保护作用及部分机制研究。方法:将实验按随机数字表法分为假手术组、脑缺血再灌注损伤组(MCAO组)、艾灸预处理每日1次组、艾灸预处理每日2次组4组,TTC染色及图像软件分析大鼠脑梗死体积,western-blot法检测脑组织中血红素氧合酶-1(HO-1)表达的变化。结果:MCAO组出现明显的神经功能缺失,各预处理组症状明显改善,且预处理组每日2次组优于每日1次组;假手术组脑组织均匀红色无梗死灶,MCAO组明显梗死苍白区;艾灸预处理较MCAO梗死体积均减小,且预处理每日2次组最为明显;MCAO组血浆CO浓度显著升高,各预处理组CO浓度降低,且每日2次组最显著;MCAO组HO-1蛋白表达均显著增加,各预处理组HO-1蛋白表达显著降低。结论:艾灸预处理能保护脑组织缺血再灌注损伤,其机制可能与通过下调HO-1蛋白表达,减少CO的合成和释放有关。
Objective: To observe the effects of moxibustion preconditioning on the protection effects and mechanisms on cerebral ischemia reperfusion injury in rats. Methods: The experiment was divided into control group,sham operation group,ischemia reperfusion injury group( MCAO group),Moxibustion preconditioning group 1 / d and moxibustion pretreatment of 2 / d group,the infarct volume of rats in each group were analyzed by TTC staining and image software,and using Western-blot method to detect the changes of heme oxygenase the enzyme-1( HO-1) expression in brain tissues.Results: MCAO group showed significant loss of nerve function,The symptoms of each pretreatment group were significantly improved,And pretreatment of the 2 / d group is better than the 1 / d group; The brain tissue of sham operation group was evenly and without infarction,MCAO group was significantly infarct pale area; Moxibustion pretreatment decreased the infarct volume of MCAO,And pretreatment of the 2 / d group is the most obvious; The plasma CO concentration of MCAO group was significantly increased,Reduction of CO concentration in each pretreatment group,And the 2 / d group is the most obvious; The expression of HO-1 protein in MCAO group were significantly increased,The expression of HO-1 protein was significantly lower in each pretreatment group. Conclusion: Moxibustion preconditioning can protect cerebral ischemia reperfusion injury,The mechanism may be related to the down-regulation of HO-1 protein expression and reducing the synthesis and release of CO.
