常压室温等离子体快速诱变选育单宁酶高产黑曲霉菌株 被引量:6

Screening of mutants of higher tannase activity strain of Aspergillus niger by atmospheric and room temperature plasmas

摘要 利用常压室温等离子体快速诱变技术对产单宁酶黑曲霉菌株B0201进行诱变,通过溴酚蓝平板变色圈法初筛,液态发酵产酶测定法复筛获得突变株B1401,其诱变后的菌株在液态发酵中获得17.61 U/g酶活,提高2倍;并在固态发酵中进行验证,在固态培养中获得酶活27.68 U/g,提高1.64倍。通过因素优化,得出其液态培养产酶的条件为单宁酸浓度5%,葡萄糖浓度2.5%,最适培养时间3.5 d,最适产酶生长温度28℃。 In order to obtain an industrial strain with higher tannase activity, the original strain of Aspergillus niger B0201 was mutated by atmospheric and room temperature plasmas. The methods used to screen the mutant strain with higher tannase activity is tablet hydrolysis using bromophenol blue circle method with liquid fermentation enzyme assay got the mutant strain B1401. The strain B1401 enzyme activity in the liquid fermentation is 17.61 U/g, 2 times than original strain. The enzyme activity in the solid state fermentation is 27.68 U/g, 1.64 times than original strain. In the liquid fermentation, the mutant tannic acid concentration optimal enzyme production was 5%, glucose concentration was 2.5%, optimal incubation time 3.5 d, optimum growth temperature for enzyme production was 28 ℃.
