红芪多糖对HLA-B27相关前葡萄膜炎TNF-α和IL-10的影响 被引量:8

摘要 目的探讨红芪多糖(HPS)对HLA-B27相关前葡萄膜炎患者外周血单个核细胞分泌细胞因子肿瘤坏死因子-α(TNF-α)和白细胞介素-10(IL-10)的影响。方法选择10名处于急性炎症期的HLA-B27相关前葡萄膜炎患者,常规药物治疗4周,在炎症缓解期,抽取他们的外周血,经过分离和培养得到单个核细胞。将单个核细胞平均分为4组,分别用磷酸盐缓冲液(PBS)、细菌脂多糖(LPS)、LPS+HPS、HPS处理各组单个核细胞,PBS处理组为对照组,LPS的终末浓度为1 mg·L-1,HPS的终末浓度为200 mg·L-1。在各组单个核细胞接受处理4 h、8 h、12 h、24 h后收集细胞培养上清液,酶联免疫吸附试验法(ELISA)检测其中TNF-α和IL-10的含量。结果患者单个核细胞受LPS处理后细胞培养上清液中各时点的TNF-α和IL-10含量均明显高于PBS处理组(P<0.05);HPS+LPS处理组细胞培养上清液中各时点的TNF-α含量以及前2个时点的IL-10含量高于LPS处理组(P<0.05);HPS处理组细胞培养上清液中的TNF-α和IL-10的含量与PBS处理组比较无明显差异(P>0.05)。结论 HLA-B27相关前葡萄膜炎炎症缓解期患者外周血单个核细胞进行的体外实验中,红芪多糖促进LPS刺激后的单个核细胞分泌TNF-α和IL-10,提示红芪多糖作为免疫调节剂参与炎症反应的过程。 OBJECTIVE To observe the effect of radix hedysari polysaccharide(HPS) on the production of tumor necrosis factor alpha(TNF-α) and interleukin-10(IL-10) by peripheral blood mononuclear cells from patients with HLA-B27 associate acute anterior uveitis(AAU). METHODS Mononuclear cells samples were isolated from 10 patients with HLA-B27 associate AAU during recovery course in its acute period of onset after 4 weeks medical treatment. Mononuclear cells isolated and cultured from the patients were divided into 4 groups, and were treated by phosphate buffered saline(PBS, as control group), lipopolysaccharide(LPS), HPS+LPS and HPS respectively. The final concentration of LPS is 1 mg·L-1. HPS was added 30 minutes before the adding of LPS in final concentration of 200 mg·L-1. Samples of culture supernatants were harvested at different times(4 h, 8 h, 12 h and24 h) after stimulation. Concentration of TNF-α and IL-10 in cell culture supernatant samples were measured with enzyme-linked immunosorbent assay(ELISA). RESULTS After stimulated by LPS, the concentrations of TNF-αand IL-10 in culture supernatants of the patients are significantly higher than control group(P〈0.05). The production of TNF-α and IL-10 of the LPS + HPS treatment group was higher than the LPS treatment group(P〈0.05). The production of TNF-α and IL-10 of the HPS treatment group shown no difference comparing with the control group.CONCLUSIONS In the in-vitro test of isolated peripheral blood mononuclear cells from the patients with HLA-B27 associate AAU after recovering, HPS promote the secretion of IL-10 and TNF-α produced by mononuclear cells stimulated with LPS. Test indicateds that HPS as immune modulators involveds in the process of inflammatory reaction.
