摘要
目的:对比5%TNBS与不同浓度无水乙醇联合液诱导大鼠UC模型的差异,优化大鼠溃疡性结肠炎的造模方法。方法:40只雌性SD大鼠,随机分为8组,包括1个正常组和7个模型组(n=5)。浓度分别为:模型1组:(5%TNBS 0.25mL+无水乙醇0.25mL);模型2组:(5%TNBS 0.2mL+无水乙醇0.2mL);模型3组:(5%TNBS 0.07mL+无水乙醇0.15mL+纯水0.8mL);模型4组:(5%TNBS0.3mL+无水乙醇0.2mL);模型5组:(5%TNBS0.07mL+无水乙醇0.12mL+纯水0.11mL);模型6组:(5%TNBS 0.1mL+80%乙醇0.1mL);模型7组:(5%TNBS0.09mL+无水乙醇0.06mL)。采用不同配比的混合液对各组大鼠分别进行灌肠,观察各组大鼠的死亡率以及体重的变化。造模10d后取距离肛门8cm处结肠组织进行病理切片。结果:与正常组比较,模型组各组均出现黑便症状以及结肠溃疡的病理改变,模型1、2、3组10的死亡率为100%,模型4组20%,模型5组为80%,模型6组为60%,造模7组没有死亡现象。结论:采用5%TNBS联合乙醇复制大鼠UC模型的最佳配置比例为:5%TNBS 0.09mL和无水乙醇0.06mL。
Objective:To compare the difference of 5% TNBS joint different concentrations of ethanol to induce UC model in rats,and optimize the methods inducing UC.Methods:40female SD rats were randomly divided into eight groups,including a control group and seven model group(n=5).The ethanol concentrations were shown as following:Model 1group:(5% TNBS 0.25mL+ethanol 0.25mL);model 2groups:(5% TNBS 0.2mL +ethanol 0.2mL);Model 3groups:(5% TNBS 0.07 mL + dry Ethanol 0.15 mL + water 0.8mL);Model 4group with 0.3mL 5% TNBS and0.2mL ethanol,model 5groups with 0.07 mL 5% TNBS and pure water and ethanol 0.11 mL 0.12 mL,model six groups with 0.1mL5% TNBS and 80%ethanol 0.1mL,model 7groups with 0.09mL5% TNBS and ethanol 0.06 mL.A mixture of different proportions of each group were lavaged.After modeling 10 d,mortality and changes in body weight of the rats were observed.and colon tissue at 8cm from the anus were taken for morphology.Results:Compared with normal group,melena symptoms and pathological changes of the colon ulcers were found in each model group.l Mortality were100%in No.1,2,3groups,20%in No.4group,80%in No.5group and 60%in No.6group,but no mortality in No.7groups.Conclusion:The optimal allocation of TNBS and ethanol for rat model of UC is 5% TNBS 0.09 mL and ethanol0.06 mL.
