三乳酸甘油酯对冷应激肉鸡肠道抗氧化能力和能量代谢状况的影响

摘要 本试验研究三乳酸甘油酯对冷应激肉鸡肠道的抗氧化能力及能量代谢状况的影响。试验选用150只1日龄Cobb肉鸡,随机分为3个处理组:对照组、冷应激组、乳酸甘油酯组,分别饲喂基础日粮、基础日粮、基础日粮+500mg/kg三乳酸甘油酯,每组5个重复,每个重复10只鸡。试验第1周各组温度控制在32~34℃,第2周控制在30~32℃,试验第3~6周冷应激组、乳酸甘油酯组控制在(10±2)℃,而对照组维持在(26±2)℃,湿度50%~60%。结果表明:与对照组相比,冷应激处理降低了肉鸡生长性能、十二指肠过氧化氢酶(CAT)、空肠超氧化物歧化酶(SOD)、CAT及谷胱甘肽过氧化物酶(GSH-Px)的活力(P〈0.05),同时也降低了十二指肠的三磷酸腺苷(ATP)、腺苷酸池(TAN)和能荷(EC)含量,回肠的ATP、二磷酸腺苷(ADP)和EC含量及空肠血红素加氧酶-1(HMOX-1)m RNA相对表达量(P〈0.05),但提高了十二指肠和回肠AMP/ATP比值,同时也提高了十二指肠、空肠、回肠AMP依赖的蛋白激酶-α1(AMPK-α1)m RNA相对表达量及十二指肠低氧诱导因子-1(HIF-1)m RNA相对表达量(P〈0.05);与冷应激组相比,日粮中添加500 mg/kg三乳酸甘油酯对冷应激肉鸡生长性能并无影响,但提高了冷应激肉鸡肠道SOD活力、十二指肠CAT和空肠GSH-Px活力、十二指肠和回肠ATP、EC含量、十二指肠和回肠血红素加氧酶-1(HMOX-1)m RNA及空肠HIF-1、COXⅢm RNA相对表达量(P〈0.05),同时降低了十二指肠和回肠AMP/ATP比值,十二指肠、空肠及回肠AMPK-α1m RNA相对表达量、十二指肠和回肠HIF-1 m RNA相对表达量及空肠PGC-1αm RNA相对表达量(P〈0.05);本试验结果表明,三乳酸甘油酯可提高冷应激肉鸡的肠道抗氧化能力、改善肠道的能量代谢状况,这可能与三乳酸甘油酯调控HIF-1/HMOX-1和AMPK信号通路有关。 This study was conducted to investigate the effects of lacti-glyceride on intestinal anti-oxidative capacity and energy metabolism status of cold-stressed broilers. 150 1-day-old Cobb chickens were randomly divided into three groups (control group, cold-stress group, lacti-glyceride group). Each treatment has 5 replicate pens and 10 chickens per pen. The control group and cold-stress group were fed a basal diet, whereas lacti-glyceridegroup were fed basal diet + 500 mg/kg lacti-glyceride. The cold-stressed process was followed as: first week 32-34℃, second week 30-32℃, third to sixth week 26±2℃ (the control group) and 10±2℃ (other groups). The humidity of all groups was 50%-60%. The results showed that: compared with the control group, cold-stress treatment decreased the growth performance, as well as the CAT activity in the duodenum, the SOD, CAT and GSH-Px activity in the jejunum, the levels of ATP, TAN and EC in the duodenum (P〈0.05), the levels of ATP, ADP and EC inthe ileum, and the HMOX- 1 mRNA level in the jejunum (P〈0.05), but increased the ratios of AMP/ATP in the duodenum and ileum, the AMPK-α1 mRNA levels in the duodenum, jejunum and ileum, and the HIF-lmRNA level inthe duodenum (P〈0.05). Compared with cold stress group, diet supplemented with 500 mg/kg lacti-glyceride had no effects on growth performance and increased the intestinal SODactivity, theCATactivity in the duodenum, the GSH-Px activity in the jejunum (P〈0.05), the levels of ATP and EC in the duodenum and ileum (P〈O.05), the HMOX-ImRNA levels in the duodenum and ileum, and the HIF-1 and COX m mRNA level in the jejunum (P〈 0.05), but decreased the ratios of AMP/ATP, HIF-1 mRNA levels in the duodenum and ileum, the AMPK-α mRNA levels in the duodenum, jejunum and ileum, andthePGC-1α mRNA level in the jejunum of cold-stress broilers (P〈0.05). In summary, these results demonstrated that the lacti-glyceride increased theintestinal anti-oxidative capacity and improvethe energy
