摘要
为了解脾虚便秘小鼠肠道细菌的特性,采用灌胃番泻叶水煎液7 d,控制饮食、饥饱失常8 d建立小鼠脾虚便秘模型,造模结束后提取肠道微生物总DNA,进行16S r DNA V4区基因组测序分析.结果显示,从OTU总数来看,模型组有896种,少于正常组的982种,二者有807种相同;从PCA分析结果来看,正常组分布相对集中,模型组彼此离散,且与正常组的距离较大;从生物多样性指数来看,模型组的Chao指数、ACE指数、Shannon指数均显著低于正常组(P<0.05),模型组的Simpson指数值则显著高于正常组(P<0.05);从细菌相对丰度来看,模型组的Clostridia、Coriobacteriales、Bacteroidales、Clostridiales、Coriobacteriia、Deltaproteobacteria、Desulfovibrionales相对丰度显著低于正常组(P<0.05),而Actinobacteria、Bifi dobacteriales、Erysipelotrichales相对丰度则极显著高于正常组(P<0.01).本研究表明脾虚便秘小鼠肠道细菌多样性明显减少,Actinobacteria、Bifi dobacteriales、Erysipelotrichales等是小鼠脾虚便秘相关的重要肠道细菌.(图3表3参25)
This research aimed to study the effects of building spleen-deficient constipation model on the intestinal bacterial diversity in mice. For construction of models of spleen-deficiency constipation, 6 mice were made hunger satiety disordered by being fed with senna water decoction for 7 days followed by diet restriction for 8 days. The intestinal microbes total DNA was extracted for genome sequencing analysis of 16S rDNA V4 area. The model group had 896 different species of OTU, less than the 982 species of the normal group, with 807 of them being common in the two groups. The PCA analysis showed relative concentrated distribution in the normal group but discrete distribution in the model group, with large distance from that of the former. The Chao index, ACE index and Shannon index were significantly lower in the model group than in normal group (P 〈 0.05), but the Simpson index value was significantly higher in the model group than in normal group (P 〈 0.05). The relative abundance of Coriobacteriales, Deltaproteobacteria, Clostridia, Bacteroidales, Clostridiales, Coriobacteriia and Desulfovibrionales was significantly lower in model group than in normal group (P 〈 0.05), while that of Actinobacteria, Bifidobacteriales and Erysipelotrichales was significantly higher in the model group (P 〈 0.01). The results showed that mice intestinal bacterial diversity is obviously reduced by the construction of spleen-deficient constipation model. Bifidobacteriales, Erysipelotrichales and Actinobacteria, etc. are the important intestinal bacteria related to the spleen-deficient constipation in mice.
