摘要
目的:探讨痛厌恶情绪对疼痛是否存在易化关系及其可能的机制。方法:雄性SD大鼠32只,随机分为5组:正常对照组(NORMAL组,n=6)、福尔马林组(Formalin组,n=6),生理盐水组(NS组,n=6),假颅内定位手术组(Sham组,n=6),颅内定位ACC毁损+福尔马林组(KA+Formalin组,n=8)。其中NORMAL组不做任何处理,Formalin组足底注入5%福尔马林50μl,NS组注入等量生理盐水,KA+Formalin组于前扣带皮层(ACC)定位注射毁损剂海人酸(Kainic acid)且足底注入5%福尔马林,Sham组ACC颅内定位注入生理盐水。测量各组机械超敏阈值(MWT)和热缩爪潜伏期(TWL)。Formalin组和KA+Formalin组还将在进行条件性位置回避测试(F-CPA)。除NS组外,皆取延髓头端腹内侧核群区域(RVM)及脊髓腰骶膨大,分别通过Western blotting,免疫组化和免疫荧光,检测RVM内色氨酸羟化酶(TPH),脊髓背角内5-HT和c-fos的表达。结果:与NORMAL,Sham组比较,KA+Formalin组的MWT和TWL降低,TPH,5-HT和c-fos表达增高(P<0.05)。与Formalin组比较,KA+Formalin组的MWT,TWL升高,TPH,5-HT表达降低(P<0.05),c-fos表达没有显著差异(P>0.05),与NORMAL组比较,NS组的MWT和TWL没有统计学差异。当单侧足底注入福尔马林的刺激同条件训练装置中的黑箱相匹配时,Formalin组大鼠在条件室(黑箱)中停留的时间较训练前明显缩短(P<0.05),而KA+Formalin组进行F-CPA匹配实验,大鼠滞留在黑箱中的时间和训练前并没有统计学差异(P>0.05)。结论:ACC的毁损可以影响痛厌恶情绪的产生,阻断痛厌恶情绪的产生可以缓解疼痛,其原因可能是打断了疼痛与痛厌恶情绪相互促进的正反馈循环,其机制可能与下行抑制/易化系统对疼痛的调节有关。
Objective: Explore the effect of the Pain-related aversion on pain and the possible mechanism.Methods: Thirty-two male SD ratswere randomly divided into 5 groups(n=6 in the first four group and n =8 in the last one): Normal control group(NORMAL), Formalin group(formalin), NS group(NS), sham operation group(sham), KA+ formalin group(KA+ Formalin).There was no operation in the normal group, but at the same time, 5% formalin 50 ul was injected in rats pelma of the formalin group and the equal NS was injected in the NS group. The KA acid was injected in ACC of the KA+ formalin group and the formalin was injected in rats pelma. At last,the NS was injected in ACC of the sham group. We measured mechanical withdrawal threshold and thermal withdrawal latency in the each group. Therefore, the formalin group and KA+ formalin group also have been tested by conditional place avoidance(F-CPA). Except the NS group, some of important materials(TPH, 5-HT) in descending facilitatory/inhibition system and the c-Fos expression in spinal cord were detected in the rest of groups. Results: MTW and TWL were significantly decreased in KA+ formalin group, and the expression of TPH, 5-HT, C-fos were increased compared with those in NORMAL and sham group(P〈0.05). Compared with formalin group, MTW and TWL were increased and the expression of TPH, 5-HT decreased(P〈0.05), but the expression of c-fos have no significant change. When the stimulus which the formalin was injected in the unilateral pelma was matched with black box in the place conditioning apparatus, the residence time of the formalin group in black box was decreased(P〈0.05). Compared with the above groups, it has no significant change to residence time of black box in the KA+ formalin group(P〈0.05). Conclusion: When we destroy the ACC, it can influence the generation of the pain-related aversion and even block its formation. The pain will be relieved. The reason may be related to block the positive feedb
