摘要
目的:观察加味苓桂术甘汤对大鼠肾纤维化模型的影响,并探讨其可能的作用机制。方法:将40只Wistar雄性大鼠随机分为4组:正常对照组、模型对照组、低剂量加味苓桂术甘汤组与高剂量加味苓桂术甘汤组。按200mg/(kg·d)腺嘌呤灌胃建立大鼠肾纤维化模型。28d后采集血液标本,检测大鼠血清BUN、Cr水平的变化;留取肾组织,用于HE染色观察肾组织病理改变及免疫组化方法检测肾组织TGF-β1的表达。结果:模型对照组血清BUN、Cr、肾组织TGF-β1较正常对照组明显升高,加味苓桂术甘汤各组上述指标较模型对照组明显下降。结论:加味苓桂术甘汤可保护肾功能,通过下调TGF-β1表达延缓肾纤维化进展。
Objective To observe the effects of Jiawei Lingguizhugan decoction in renal fibrosis rats and explore its possible mech-anism.Methods 40 Wistar rats were randomly divided into four groups,namely the control group,the model group,the low -dose Jiawei Lingguizhugan decoction group and the high -dose Jiawei Lingguizhugan decoction group.The models of renal fibrosis were established by intragastric administration of adenine200mg/(kg·d).The blood samples were collected after 28 days.The changes of blood urea nitrogen and creatinine were detected.The kidney pathological changes were observed by light microscopy with HE -staining.The expression of TGF -β1 was determined by immunohistochemical method.Results Blood urea nitrogen,creatinine and TGF-β1 of the model group were markedly increased than that of the control group.The above indicators were lower in the low -dose and high -dose Jiawei Lingguizhugan decoction groups than those in the model group.Conclusions Jiawei Lingguizhugan decoction could protect the kidney function and postpone renal fibrosis by decreasing the expression of TGF -β1 in renal tissues.
