摘要
采用乙醇注入法制备顺铂脂质体.分别采用凝胶色谱法、超速离心法及中空纤维超滤法分离载药脂质体和游离药物,并用电感耦合等离子体-发射光谱(ICP-OES)测定其中的铂含量,计算顺铂脂质体的包封率.实验结果显示ICPOES法测定铂浓度在1 100μg·m L^(-1)的范围内线性关系良好(r=0.999 9);高、中、低3种浓度的日内和日间精密度试验的RSD均小于1%;3种包封率测定方法中中空纤维超滤法测得的包封率最高(98%)且最快,超速离心法测得的包封率最低(67%).
The cisplatin liposomes were prepared by ethanol injection. The liposomes and free drug were separated by size exclusion chromatography, ultracentrifugation and hollow fiber ultrafiltration, then Pt concentration were determined by Inductively Coupled Plasma Optical Emission Spectrometry( ICP-OES) ,and then the entrapment efficiency of the liposomes was calculated. The Results shows that the calibration curve was linear(r=0. 999 9) with in the range of 1—100 μg·mL-1 for Pt determined by ICP-OES. The RSDs of the intra-day and liter-day were less than 1%. Among the three methods,the entrapment efficiency determined by hollow fiber ultrafiltration was highest ( 98%) and the entrapment efficiency determined by ultracentrifugation was the lowest(67%) .
