摘要
药物性肝损伤一直是创新药物研发关注的重点和难点,也是引发药物物研发失败和退市的主要原因之一。鉴于目前药物毒理研究的最新进展,基于毒性通路的药物安全评价和生物标志物的寻找和验证已逐渐成为近年来的研究热点。本文从药物肝损伤相关的毒性通路,生物标志物,相关的分子起始事件等几个方面进行了综述,以希为找到降低药物肝损伤和能够提前预警药物肝毒性筛选的方式提供些参考。
Drug-induced liver injury has been the focus and difficulty of the research and development of innovative drugs. It is also one of the main reasons for the failure of drug development. With the progress in the research of drug toxicity,drug safety evaluation based on toxicity pathway and the search and validation of biomarkers have gradually become a hot research topic in recent years. In this paper,we reviewed the possible related toxicity pathways,biomarkers,and related molecular initiation events. In order to find ways to reduce drug-induced liver injury and provide a reference for the early screening of drug-induced liver toxicity.
