酮咯酸氨丁三醇对大鼠脑缺血再灌注损伤的神经保护作用 被引量:3

Neuroprotective effect of ketorolac tromethamine on the cerebral ischemia - reperfusion injury in rats

摘要 目的观察酮咯酸氨丁三醇对大鼠局灶性脑缺血再灌注损伤的脑保护作用。方法采用改良线拴法阻断大鼠大脑中动脉2h,再灌注24h制备大脑中动脉阻塞模型(MCAO),将48只sD雄性大鼠随机分为假手术组、模型组和酮咯酸氨丁三醇组(4mg/kg),对其进行神经功能评分、2,3,5-氯化三苯基四氮唑(TTC)染色测定脑梗死面积并检测大鼠再灌注后脑组织中细胞因子[白细胞介素(IL)-1β、IL-6、肿瘤坏死因子-α(TNF-α)]的表达。结果神经功能评分和脑梗死面积:与模型组[(2.78±0.93)分,(39.96±15.12)%]比较,酮咯酸氨丁三醇组[(1.59±0.61)分,(25.04±8.14)%]神经功能评分和脑梗死面积降低(P〈0.05);细胞因子(IL-1β、IL-6、TNF-α)的表达:与假手术组[(0.128±0.006)、(0.105±0.312)、(1.498±0.701)μg/L]比较,模型组[(0.371±0.026)、(0.217±0.056)、(2.762+0.141)μg/L]含量增高(P〈0.05);与模型组比较,酮咯酸氨丁三醇组[(0.164±0.005)、(0.147±0.028)、(1.873±0.207)μg/L]含量降低(P〈0.05)。结论酮咯酸氨丁三醇对大鼠脑缺血再灌注损伤具有脑保护作用。 Objective To study the neuroprotective effect of ketorolac tromethamine on the cere- bral ischemia- reperfusion injury in rats. Methods Focal cerebral ischemia/reperfusion model was con- ducted using improved suture - occluded method by occluding middle cerebral artery for 2 h followed by 24 h of reperfusion. Adult male SD rats were randomly divided into three groups (u = 16 each): sham group, model group and ketorolac tromethamine group (4 mg/kg). The neurological function was scored, infarction area was measured by 2, 3, 5 - triphenyltetrazolium chloridec ( TYC ) staining, and the content of brain tissue cytokine [ interleukin (IL) -1β, IL-6, tumor necrosis factor-ct (TNF-α)] was deter- mined after reperfusion. Results Behavioral scores and the infarct volume: compared with the model group [ 2. 78 ± 0. 93, (39.96 ± 15. 12) % ], ketorolae tromethamine group [ 1. 59 ± 0. 61, (25.04 ± 8. 14) % ] ruduced behavioral scores and the infarct volume ; cytokine ( IL - 1β, IL - 6, TNF - α) expression : com- pared with the sham group [ (0. 128 ±0. 006), (0. 105 ±0. 312), ( 1. 498 ±0. 701 ) μg/L] ,the model group [ (0. 371 ±0. 026), (0. 217 ±0. 056), (2. 762 ± 0. 141 ) pg/L] inseared the content; compared with the model group, ketorolac tromethamine group [ ( 0. 164 ± 0. 005 ), ( 0. 147 ± 0. 028 ), ( 1. 873 ± 0. 207 ) μg/L] lowered the content ( P 〈 0. 05 ). Conclusion Ketorolac tromethamine has neuroprotective effect on cerebral ischemia- reperfusion injury in rats.
