摘要
【目的】比较白花蛇舌草中各活性成分抑制肿瘤细胞CT-26增殖的效果。【方法】小鼠结肠癌细胞CT-26常规培养,不同浓度的山奈酚、熊果酸、齐墩果酸、阿魏酸、豆甾醇、三十一烷、芦丁干预,细胞计数试剂盒8(cell counting kit-8,CCK-8)法检测并描绘细胞生长曲线,倒置显微镜观细胞形态变化,流式细胞仪检测药物对细胞凋亡及细胞周期的影响。【结果】药物干预后,CT-26的细胞密度明显减小,单个细胞体积缩小呈圆形,最终悬浮脱落而死亡;细胞增殖受到抑制,并呈现出量效关系。【结论】白花蛇舌草各活性单体能显著抑制小鼠结肠癌细胞CT-26的增殖,诱导细胞凋亡。
【Objective】To compare the effects of active ingredients in Oldenlandia diffusa on suppressing the proliferation of colon cancer cells(CT-26).【Methods】Mouse CT-26 cells were cultured routinely and treated with different concentrations of kaempferol,ursolic acid, oleanolic acid, ferulic acid, stigmasterol, hentriacontane and rutin. Cell Counting Kit-8 was used to determine cell survival rate, then cell growth curve was drawn. Cell morphological changes were detected with inverted microscope. The influence of drugs on cell apoptosis and cell cycle was determined with flow cytometry.【Results】After the intervention of drugs, the density of CT-26 cells obviously minished; the single cell shrank, appeared round, eventually suspended off and die; cell proliferation was suppressed in a dose- effect relationship.【Conclusion】Each active ingredient in Oldenlandia diffusa can significantly suppress the proliferation of mouse CT-26 cells and induce cell apoptosis.
