七氟醚后处理对糖尿病大鼠心肌缺血再灌注损伤线粒体融合蛋白1表达的影响

摘要 目的:探讨七氟醚后处理对糖尿病大鼠心肌缺血再灌注损伤时线粒体融合蛋白1(Mfn1)表达的影响。方法:健康成年SD大鼠60只,体重200-240 g,采用随机数字表法将其分成假手术组(S组)、糖尿病假手术组(DS组)、糖尿病缺血再灌注组(DI/R组)和糖尿病七氟醚后处理组(DSev组),每组15只。采用腹腔一次性注射链脲佐菌素65 mg/kg的方法制备糖尿病模型。采用结扎左冠状动脉前降支制备心肌缺血再灌注模型,S组和DS组只穿线不结扎,DI/R和DSev组结扎左冠状动脉的前降支30 min,再灌注120 min,DSev组再灌注前1 min吸2.5%的七氟醚5min,再灌注结束时取大鼠心肌组织。采用2,3,5-氯化三苯基四氮唑双重染色法(TTC)测定心肌梗死面积,TUNEL法测定凋亡指数(AI),免疫组化法测定各组Mfn1的表达。结果:与S组相比,DS组凋亡指数增加,Mfn1的表达下调(P〈0.05),心肌梗死范围差异无统计学意义(P〉0.05),DI/R组和DSev组心肌梗死面积和AI增加,Mfn1的表达下调(P〈0.05)。与DS组相比,DI/R组和DSev组心肌梗死范围和AI增加,Mfn1的表达下调(P〈0.05)。与DI/R组相比,DSev组心肌梗死面积、AI及Mfn1的表达差异无统计学意义(P〉0.05)。结论:糖尿病大鼠心肌Mfn1的表达较正常大鼠下调,糖尿病缺血再灌注损伤可进一步使Mfn1的表达下调,七氟醚后处理对糖尿病大鼠心肌缺血再灌注损伤线粒体Mfn1的表达无明显影响。 Objective: Toexplore the effects of sevoflurane postconditioning on the express of mitofusion- 1 in diabetes rats with ischemia reperfusion injury Methods: Sixty healthy adult male Sprague- Dawley rats,weighing 200 - 240 g,were randomly divided into 4 groups( n = 15 each) : normalsham operation group( S group),diabetic sham operation group( DS group),diabeticischemia- reperfusion group( DI / R group) and diabeticsevoflurane postconditioning group( DSev group). The model of diabetes mellitus was induced by intraperitoneal streptozotocin 65 mg / kg. The model of myocardial ischemia / reperfusion was produced by occlusion of the anterior descending branch of left coronary artery for 30 minutes followed by 120 minutes reperfusion in rats. Threading but not ligation was induced in S group and DS group. The anterior descending branch of left coronary artery was occluded for 30 minutes andreperfused for 120 minutes in DI / R group and DSev group. 2. 5% sevoflurane was inhaled for 5 minutes starts from 29 min of ischemia in DSev group. After 120 minutes of reperfusion,rats were sacrificed and cardiac tissues were obtained. Myocardial infarct size was measured by TTC staining. Cardiomyocyte apoptosis was determined by TUNEL. The expression of Mfn1 protein levels was determined by immunohistochemistry. Results: Compared with S group,the cardiomyocyte apoptosis index was increased,the express of Mfn1 protein was decreased( P〉0. 05),the myocardial infarct size has no significant difference in DS group( P〈0. 05). The myocardial infarct size and the myocardiomyocyte apoptosis index was significant increasedthe expression of Mfn1 protein was obviously decrease,in DI / R groupand DSev group( P〈0. 05).Compared with DS group,the myocardial infarct size and the myocardiomyocyte apoptosis index was significant increased,the express of Mfn1 protein was obviously decreased in DI / R group and DSev group( P〈0. 05).
