摘要
目的观察真核细胞起始因子4E(e IF4E)、血管内皮生长因子(VEGF)在人脑星形细胞瘤组织的表达及意义。方法采用免疫组化Elivision TMplus两步法和Real time PCR法检测75例脑星形细胞瘤组织和40例正常脑组织中e IF4E、VEGF蛋白和mRNA表达情况,分析二者表达与微血管密度(MVD)的相关性。结果正常脑组织e IF4E、VEGF蛋白阳性表达率分别为12.50%(5/40)、7.50%(3/40),脑星形细胞瘤组织e IF4E、VEGF蛋白阳性表达率分别为69.33%(52/75)、64.00%(48/75),脑星形细胞瘤组织e IF4E、VEGF阳性表达率显著高于正常脑组织,且二者mRNA表达水平也显著高于正常脑组织,差异有统计学意义(P〈0.05)。脑星形细胞瘤Ⅱ~Ⅳ级组,e IF4E、VEGF阳性表达率逐渐增加,分别为40.00%(10/25)、75.86%(22/29)、95.24%(20/21)和32.00%(8/25)、62.07%(18/29)、80.95%(17/21),差异有统计学意义(P〈0.05);且有转移组e IF4E、VEGF阳性表达率显著高于无转移组(P〈0.05)。e IF4E、VEGF表达呈正相关性(r=0.768,P〈0.05)。随着脑星形细胞瘤组织e IF4E、VEGF表达强度增高,MVD值逐渐增加,差异有统计学意义(P〈0.05)。结论 e IF4E、VEGF在脑星形细胞瘤组织异常高表达,随着肿瘤恶性程度增高而逐渐升高,且与微血管密度密切相关。e IF4E或VEGF有可能成为e IF4E、VEGF治疗的潜在靶点。
Objective To investigate the expressions and significance of eukaryotic initiation factor 4E( eI F4E) and vascular endothelial growth factor( VEGF) in human astrocytoma.Methods The expression levels of eI F4 E and VEGF were detected by immunohistochemical assay and Real time-PCR in 75 cases of human astrocytoma tissues and 40 cases of normal brain tissues.The correlation between the expressions of eI F4 E,VEGF and tumor microvascular density( MVD) was analyzed.Results The positive expression rates of eI F4 E and VEGF protein were 69.33%( 52 /75) and 64.00%( 48 /75)in astrocytoma tissues,which were obviously higher than those in normal brain tissues that were 12.50%( 5 /40) and 7.50%( 3 /40),respectively,moreover,the expression levels of eI F4 E and VEGF mRNA in astrocytoma tissues were significantlyhigher than those in normal brain tissues( P〈0.05).From brain astrocytoma of grade Ⅱ to brain astrocytoma of grade Ⅳ,the positive expression rates of eI F4 E and VEGF were gradually increased,which were 40.00%( 10 /25),75.86%( 22 /29),significantly higher than those in non tumor metastasis group( P〈0.05).Besides theexpression of eI F4 E was positively correlated to that of VEGF 95.24%( 20 /21) VS 32.00%( 8 /25),62.07%( 18 /29),80.95%( 17 /21),respectively,there were significant differences among them( P〈0.05).Furthermore,the positive expression rates of eI F4 E and VEGF in tumor metastasis group were( r = 0.768,P〈0.05).With the increase of expression levels of eI F4 E and VEGF,MVD was gradually increased( P〈0.05).Conclusion The high-expressions of eI F4 E and VEGF appear in brain astrocytoma tissues,with the increase of malignant degree of tumor,the expression levels are gradually increased,moreover,which are closely correlated to MVD.The eI F4 E or VEGF may become potential target point in oncotherapy.
