mTOR和eIF4E蛋白在大肠癌组织中的表达及临床意义

摘要 目的研究eIF4E和mTOR在结直肠癌中的表达及其临床病理关系。方法外科手术中获取大肠癌组织及癌旁组织各60例,采用RT-PCR及ELISA检测大肠癌组织及癌旁组织中mTOR和eIF4E基因的表达。结果大肠癌组织中mTOR和eIF4E基因的表达明显高于癌旁正常组织,差异有统计学意义(P<0.05)。mTOR和eIF4E基因表达与性别、年龄、临床TNM分期差异无统计学意义(P>0.05),在不同肿瘤分化程度以及有无淋巴结转移上差异有统计学意义(P<0.05)。相关性分析显示,mTOR和eIF4E在大肠癌中的表达呈显著正相关(P<0.05)。结论 mTOR和eIF4E基因在大肠癌中过度表达,联合检测两者在大肠癌中的表达可能有助于恶性程度的评估与预后判断。 Objective Research the relationship between the expression of elF4 E and mTOR and the clinical and pathological features in colorectal cancer tissue. Methods collect per 60 cases colorectal cancer tissue and normal tissue adjacent cancer when surgery, using reverse transcriptase polymerase chain reaction technology and enzyme league immune method to detect the changes of elF4 E and mTOR gene. Results The expression of elF4 E and mTOR in colorectal cancer tissues is obviously higher than normal tissue adjacent to cancer, the difference was statistically significant (P〈0.05). The expression of elF4 E and roTOR in colorectal cancer was related to degree of cell differentiation tumor and lymph node metastsis (P〈0.05), but not to age, sex, TNM Stages (P〉0.05). There was positive correlation between elF4 E protein and mTOR protein expression in colorectal cancer tissue (P〈0.05). Conclusion The expression of elF4 E and mTOR gene is excessive in colorectal cancer; United detection the expression of elF4 E and mTOR gene colorectal cancer may help to evaluate the malignant degree and the judgment about prognosis.
