摘要
目的通过多指标综合加权评分法优选大黄药材中蒽醌类成分提取工艺。方法 HPLC法测定大黄药材中芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚5个蒽醌类成分游离含量及总蒽醌含量,采用正交试验法,以乙醇浓度、乙醇用量、提取时间和提取次数为考察因素,以总蒽醌提取率为考察指标,综合加权评分法进行数据分析。结果最佳工艺参数为每次加5倍量75%乙醇回流提取0.5 h,提取5次。结论优选的工艺简单、可行,可用于大黄药材中蒽醌类成分的提取。
Objective To optimize the extraction of anthraquinones from Rhei Radix et Rhizoma by multi-index comprehensive weighted score evaluation. Methods The content of 5 free anthraquinones(aloe-emodin, rhein, emodin, chrysophanol, and physcion) and total anthraquinones in Rhei Radix et Rhizoma were determined by HPLC. Orthogonal test was used to optimize the extraction process, with ethanol concentration, solvent ratio, extraction time, and number of extractions as the detecting factors and the extraction rate of total anthraquinones as examining index. The data were analyzed by multi-index comprehensive weighted scoring evaluation. Results The best extraction conditions were 5 times of 75% ethanol reflux for 5 extractions lasting 0.5 h each time. Conclusion The extraction process is simple and appropriate, and can be used for the extraction of anthraquinones from Rhei Radix et Rhizoma.
