摘要
目的探讨CO_2气腹与胃癌细胞在裸鼠腹腔增殖、转移及其机理。方法胃癌细胞株SGC-7901注射到30只裸鼠腹腔建立腹腔胃癌脱落细胞模型,随机分为麻醉组、开腹组、CO_2气腹0.5 h组和CO_2气腹1.0 h组。8周后观察肿瘤生长情况,反转录-聚合酶链反应(RT-PCR)和Western-Blot检测各瘤体组织中血管内皮生长因子(VEGF)、大鼠缺氧诱导因子-1a(HIF-1a)、天冬氨酸蛋白水解酶3(Caspase-3)、BCL-2的mRNA及蛋白表达水平。结果 CO_2组肿瘤重量明显高于麻醉组及开腹组,CO_2气腹1 h组高于CO_2气腹0.5 h组,开腹组与对照组比较,差异无统计学意义(P>0.05);RT-PCR与Western blot检测结果显示,VEGF,HIF-1a,BCL-2在气腹组中的表达高于对照组及开腹组(P<0.05),1 h组表达高于0.5 h组(P<0.05),开腹组大于对照组(P<0.05),气腹组Caspase-3表达低于对照组及开腹组(P<0.05),1 h组表达高于0.5 h组(P>0.05),开腹组与对照组比较,差异无统计学意义(P>0.05);结论 CO_2气腹能增强并随着时间增加腹腔脱落胃癌细胞黏附、生长、增殖的能力。
Objective To discuss the proliferation and metastasis of gastric cancer cells in the CO2 pneumoperitoneum environment and its mechanism. Methods A peritoneal transplantation model of stomach carcinoma was established by using the SGC7901 cell hne. The mice bearing tumors were randomly divided into four groups:simple anesthesia,laparotomy,O. 5h of CO2 insufflation, and 1 h of CO2 insufflation. Eight weeks later, we collected the tumor and weight total weight of implanted nodules. Then, the levels of VEGF, HIF-I a, Caspase-3 and BCL-2 were measured by RT-PCR and Western blot, respectively. Results The total weight of tumors in the CO2 insufllation group were all significantly higher than those in the anesthesia-group and laparotomy group ( P 〈 0.05 ). There were no significant differences among the groups between the anesthesia group and laparotomy group ( P 〉 0. 05 ). Comparing with the anesthesia group and laparotomy group, the levels of VEGF, HIF-1 and BCL-2 were increase in the CO2 insufltation groups ( P 〈 0.05 ). However, the level of Caspase-3 was much lower in the CO2 insufflation groups, compare with anesthesia group and laparotomy group ( P 〈 0. 05 ). Conclusion CO2 pneumoperitoneum significantly facilitate the proliferation and metastasis of gastric cancer cells in peritoneal transplantation model.
