摘要
目的:探讨中药清毒栓对宫颈癌Si Ha细胞增殖的影响及其诱导凋亡作用的可能机制。方法:设立空白对照组,清毒栓高、中、低剂量组(0.16、0.04、0.01 g/ml)及保妇康栓组(0.017 5 g/ml),分别以不同药液作用于宫颈癌Si Ha细胞24 h、48 h、72 h,采用CCK-8(Cell Counting Kit-8)法检测细胞增殖情况,采用线粒体膜电位法检测细胞凋亡情况。结果:不同剂量的清毒栓作用24 h、48 h、72 h时对宫颈癌Si Ha细胞的增殖均具有一定的抑制作用,其中以清毒栓高剂量组在48 h时作用最为明显。不同剂量的清毒栓均可引起宫颈癌Si Ha细胞线粒体膜电位的改变,进而引起不同程度的细胞凋亡;经统计学分析显示,清毒栓各剂量组、保妇康栓组的凋亡率与空白对照组比较,差异均有统计学意义(P<0.05),清毒栓低、中剂量组与清毒栓高剂量组比较,差异亦有统计学意义(P<0.05)。结论:高剂量的中药清毒栓在48 h时对宫颈癌Si Ha细胞增殖的抑制作用最为明显,其作用机制可能是通过使线粒体膜电位降低来促进肿瘤细胞的凋亡。
Objective: To study the effect of"Qingdu Suppository"on the proliferation of cervical cancer Si Ha cells and its mechanisms of inducing apoptosis. Methods: Five groups including blank control group,"Qingdu Suppository "high-,middle-,low-dose groups( 0. 16,0. 04,0. 01 g / ml) and"Baofukang Suppository"group( 0. 017 5 g / ml) were established.The Si Ha cells were treated by different drugs for 24 h,48 h and 72 h. The cell proliferation was detected by Cell Counting Kit-8( CCK-8) assay and the cell apoptosis was detected by mitochondrial membrane potential assay. Results: "Qingdu Suppository"of different doses can inhibit the proliferation of Si Ha cells at 24 h,48 h and 72 h,and the high-dose"Qingdu Suppository"has the most inhibiting effect at 48 h. "Qingdu Suppository " of different doses can lead to the change of mitochondrial membrane potential and induce the cell apoptosis; the apoptosis rates of "Qingdu Suppository " groups and"Baofukang Suppository"group were higher than that of the blank control group( P〈0. 05) and the apoptosis rates of"Qingdu Suppository"low-and middle-dose groups were lower than that of the "Qingdu Suppository"high-dose group( P〈0. 05).Conclusion: "Qingdu Suppository"of high-dose has the most inhibiting effect on the proliferation of cervical cancer Si Ha cells and its mechanisms may be through the reduction of mitochondrial membrane potential to induce the tumor cell apoptosis.
