摘要
目的:观察丹参酮Ⅱ-A(TsⅡ-A)对大鼠大鼠成骨细胞(OB)中骨形成蛋白-2(BMP-2)mRNA表达的影响。方法:培养原代大鼠OB并传代接种后采用Cell Counting Kit-8试剂盒比色法检测5×10^(-3)、5×10^(-2)以及5×10^(-1)mg/L 3个浓度的TsⅡ-A(统称实验组)和对照组(0 mg/L TsⅡ-A)在第1、3、5、7天对OB的促增殖作用;RT-q PCR检测各组OB BMP-2 mRNA的表达。结果:不同浓度TsⅡ-A对OB均有促增殖的作用(P<0.05),其中5×10^(-2)mg/L浓度组促增殖作用最大;在各时间段中,组间比较OB BMP-2 mRNA表达量,实验组均高于对照组(P<0.05);各实验组组内比较,OB BMP-2 mRNA表达趋势为第7天>第5天>第3天>第1天(P<0.05)。结论:TSⅡ-A在一定的作用时间内可促进OB增殖,并上调OB中BMP-2 mRNA表达水平。
Objective: To investigate the effects of Tanshinone Ⅱ-A on expression of the bone morphogenetic protein 2( BMP-2) mRNA of rat osteoblasts. Methods: the original generation of rat osteoblasts were cultivated and passaged,and Tanshinone Ⅱ-A was supplemented into the culture medium of osteoblast at 5 × 10^-3mg / L,5 × 10^-2mg / L,5 ×10^-1mg / L respectively. Cell Counting Kit-8 Kit colorimetry were used to detect the proliferation of osteoblast on the 1^(st),3^(rd),5^(th),7^(th) day,the expression of BMP-2 mRNA was checked by RT-q PCR. Results: All concentrations of Tanshinone Ⅱ-A were promoting proliferation of osteoblast( P〈0. 05),which 5 × 10^-2mg / L demonstrated the greatest effect. The expression of BMP-2 mRNA of experimental group was higher than control group in each period through comparison( P〈0. 05),while the intra-group comparison displayed that the expression of BMP-2 mRNA showed an increasing trend with time went by( P〈0. 05). Conclusions: TanshinoneⅡ-A promoted the proliferation of osteoblasts in a certain time significantly,and which also could increase the expression level of BMP-2 mRNA.
