摘要
为了研究猪白细胞介素18是否增强猪细小病毒(PPV)灭活疫苗在猪体免疫效果,分别将猪白细胞介素18与自制猪细小病毒灭活疫苗和猪细小病毒灭活疫苗联合免疫仔猪,并设猪白介素18和生理盐水对照组。免疫后分别用间接ELISA方法和流式细胞技术检测仔猪抗体水平变化和仔猪外周血T淋巴细胞亚群动态变化。结果显示,免疫后猪白细胞介素18联合灭活疫苗组抗体水平与无猪白介素18灭活疫苗组相似,但免疫后猪白介素18联合PPV灭活疫苗组的CD3^+、CD3^+CD4^+、CD3^+CD8^+T淋巴细胞所占比值均高于相应的PPV灭活疫苗组,但差异不显著,且CD4^+/CD8^+在正常范围内。研究证实猪白介素18主要增强PPV油乳剂灭活疫苗的细胞免疫应答。
The objective of this study is to characterize the enhancing immune responses to inactivated porcine parvovirus( PPV) oil emulsion vaccine by co-inoculating porcine interleukin 18( IL-18) in porcine. In this study,the immunogenicity of PPV vaccine and the immuno-regulatory activities of IL-18 were investigated. The inactivated PPV vaccines with or without IL-18 were inoculated into piglet by subcutaneous injection. Then humoral and cellular immune responses were evaluated by indirect enzyme-linked immunosorbent assays( ELISA) and fluorescence-activated cell sorter analyses( FACS).The results demonstrated that after immunity,the swine IL-18 joint inactivated vaccine group antibody levels were similar to inactivated vaccine without IL-18,and the T lymphocyte ratio of swine IL-18 joint PPV inactivated vaccine group CD3~+,CD3~+CD4~+,CD3~+CD8~+were higher than PPV inactivated vaccine group,but the difference was not significant. In conclusion,these results suggest that IL-18 possess better cellular immune-enhancing capability and can be exploited into an effective immune-adju-vant for inactivated vaccines.
