针刺对血管性痴呆大鼠海马Ref-1上下游调控分子的影响 被引量:8

Effects of acupuncture on hippocampal Ref-1 upstream and downstream molecular in rats with vascular dementia

摘要 目的:观察针刺对血管性痴呆大鼠海马Ref-1上下游调控分子的影响,探讨针刺基于Ref-1信号通路的抗氧化机制。方法:Wistar大鼠82只,随机分为假手术组、模型组、针刺组和非穴组。双侧颈总动脉阻断法复制血管性痴呆大鼠模型,造模后第3天针刺,针刺治疗结束后,采用Morris水迷宫检测大鼠认知功能;尼氏染色检测大鼠海马神经元凋亡情况;Western blot技术检测大鼠海马磷酸化CKⅡ蛋白表达水平;免疫组化技术检测大鼠海马HIF-1α蛋白表达情况。结果:与假手术组比较,造模后大鼠认知功能受损,海马区神经元凋亡,磷酸化CKⅡ水平显著下降(P<0.05),HIF-1α表达水平显著升高(P<0.05)。针刺组大鼠学习记忆能力及神经元凋亡情况显著改善,磷酸化CKⅡ水平显著升高(P<0.05),HIF-1α表达水平显著降低(P<0.05),且改善程度显著优于非穴组(P<0.05)。结论:针刺通过提高海马Ref-1上游CKⅡ的磷酸化表达水平,降低海马Ref-1下游HIF-1α表达水平,发挥抗氧化作用,抑制神经元细胞凋亡,改善学习记忆能力。 Objective: To observe the effect of acupuncture on hippocampal Ref-1 upstream and downstream molecular in rats with vascular dementia and explore the antioxidant mechanism of acupuncture based on Ref-1 signaling pathways. Methods: Eighty-two male Wistar rats were randomly divided into four groups: sham-operation group, model group, acupuncture group and placebo-acupuncture group. Vascular dementia model was established using bilateral carotid artery occlusion. Acupuncture was performed at three days after surgery, once daily for 14 days(with a rest every 7 days). Morris water maze and Nissl's staining were used to test the cognitive function and neuron apoptosis respectively. The protein expressions of hippocampal phosphorylates CKI were tested by Western blot. HIF-1α expressions in hippocampus were assessed by immunohistochemical method. Results: Compare with the sham-operation group, the vascular dementia model showed the impaired cognitive function, neuron apoptosis, decreased phosphorylates CKI protein and enhanced HIF-1α in hippocampus, which were reversed by the treatment of acupuncture(P<0.05). Acupuncture improued cognitive functions, inhibit neuron apoptosis, and reverse the decrased of phosphorylaties and increased of HIF-1α expression(P<0.05). These effects were not found in the placebo-acupuncture group. Conclusion: The antioxidant effect of acupuncture may be achieved by increasing the phosphorylates CKI levels in Ref-1 upstream and decreasing the HIF-1α expression in Ref-1 downstream of hippocampus, resulting in inhibit neuron apoptosis and improving cognitive functions.
