针刺对骨骼肌顿挫伤模型兔CaN/NFAT信号通路的影响 被引量:1

Effect on Signal Pathway of Calcineurin- NFAT in Rabbit Models with Soft Tissue Injury by Acupuncture

摘要 目的:观察针刺对兔骨骼肌钝挫伤后组织超微结构和CaN/NFAT信号通路的影响,探讨该通路在骨骼肌损伤修复中的作用机制。方法:30只家兔,雌雄各半,随机分为空白组、模型组、环孢素A(CsA)组、针刺组、针刺合并CsA组。采用重物自由落体撞击法造成急性骨骼肌钝挫伤模型。针刺组于造模24 h后采用毫针斜刺腓肠肌全肌束的方法干预,CsA组于造模前3天按每天每只兔9.25mg/kg CsA进行注射,针刺合并CsA组进行针刺合并CsA干预。采用Western blot法检测CaN和NFAT蛋白表达量。结果:电镜下针刺组肌丝总体排列整齐,肌节未见明显肿胀,Z线、M线清晰;模型组、CsA组和针刺合并CsA组均有不同程度的肌原纤维断裂、溶解,Z线、M线有不同程度的溶解断裂。与空白组相比,各组CaN蛋白表达量均显著升高(P<0.05或P<0.01),模型组、针刺组和针刺合并CsA组NFAT蛋白表达显著升高(P<0.05);与模型组相比,针刺组CaN和NFAT蛋白表达量显著升高(P<0.05);与CsA组相比,针刺组与针刺合并CsA组CaN和NFAT蛋白表达量显著升高(P<0.05)。结论:针刺可有效改善急性钝挫伤模型兔超微结构的损伤;急性软组织顿挫伤可使兔骨骼肌CaN、NFAT1蛋白表达升高;针刺可激活CaN/NFAT信号通路,促进骨骼肌的再生修复。 Objective: To observe the effect on signal pathway of Calcineurin- NFAT in rabbit models with soft tissue injury by acupuncture,so as to reveal its mechanism. Methods: Thirty rabbits were randomized into a blank group,a model group,a CsA group,a needling group and a needling combined with CsA group,with 6 rabbits in each group. The acute STI modles were established by heavy objects in free fall for consecutive shocks.The needling group was given oblique insertion of the needles at Ashi point. CsA group was given intraperitioneal injection of 9. 25 mg / kg CsA and needling combined with CsA group was given both needling and CsA after24 h of modeling. There was no intervention in the model group and blank group. Use electron microscope to observe the changes of the ultrastructure of skeletal muscle structure,and the Calcineurin and NFAT protein expression of Gastrocnemius musle was detected by western blot. Results: Needling group under the electron microscope had not seen obvious swelling sarcomere. Z line and M line were clear; Model group,CsA group and needling combined with CsA gro- up all had different degrees of myofibril fracture and dissolution. Z line and M line had different degrees of dissolved fracture. Compared with the blank group,in other groups CaN quantities of protein expression were significantly increased( P〈0. 05 or P〈0. 01),and in model group,CsA group,needling group and needling combined with CsA group each amount of NFAT protein expression was significantly increased( P〈0. 05); compared with model group,CaN and NFAT markedly increased in the amount of protein expression for the needling group( P〈0. 05); compared with CsA group,CaN and NFAT markedly increased in the amount of protein expression for the needling group and needling combined with CsA group( P〈0. 05).Conclusion: Acupuncture can effectively improve the injury of the ultrastructure of acute blunt trauma models;acute soft tissue contusion can increase the rabbit skeletal muscle CaN and NFAT1 protein
