基于UPLC/QTOF-MS技术的补骨脂水提物对大鼠血浆代谢组学研究 被引量:7

Metabonomics study of aqueous extract of Fructus Psoraleae on serum of rats based on UPLC / QTOF-MS

摘要 目的:研究补骨脂水提物对大鼠血清内源性代谢产物的影响,并从中发现相关代谢标志物。方法:雄性SD大鼠每天1次分别给予补骨脂水提物0.54,1.08和1.62 g/kg,连续给药2周,末次给药后,禁食24h,采集血液,应用超高效液相色谱/四级杆飞行时间质谱(UPLC-Q-TOF-MS)分析样品,Mass Lynx采集并处理数据,采用偏最小二乘判别分析(PLS-DA)方法,分析各组大鼠血清内源性代谢产物的差别,通过正交偏最小二乘法(OPLS-DA)、变量重要程度(VIP)筛选潜在生物标志物并测定其相对含量。结果:与空白组相比,各给药组吲哚、L-苯丙氨酸和植物鞘氨醇含量呈明显上升,乳酸和芽子碱甲酯含量呈明显下降(P<0.05,P<0.01)。结论:补骨脂水提物给药2周后,大鼠血清内源性物质发生显著改变,补骨脂水提物发挥药效或毒效的机制可能是通过调节氨基酸代谢、磷脂代谢以及糖酵解过程来实现的。 Objective: To study the effect of aqueous extract of Fructus Psoraleae on the endogenous metabolites in rat's serum and find the relative endogenous marker. Methods: Male SD rats were orally administrated with aqueous extract of Fructus Psoraleae at the dose of 0. 54,1. 08 and 1. 62 g / kg,respectively,once a day for two consecutive weeks. Serum samples were collected 24 h after the last administration. Data were acquired with the Mass Lynx software based on ultra-performance liquid chromatography / quadrupole time of flight mass spectrometry( UPLC-Q-TOF-MS),the partial least squares-discriminant analysis( PLS-DA) were used to analysis the difference of endogenous metabolites in different groups,then potential biomarkers were found through orthogonal partial least-squares-discriminant analysis( OPLS-DA) and variable importance in the projection( VIP) and their relative intensity were determined. Results: Compared with the control group,the relative contents of indole,L-Phenylalanine and Phytosphingosine in each group were increased in a dose-dependent manner( P 0. 05,P 0.01),the relative contents of Lactic acid and Ecgonine methyl ester were decreased in a dose-dependent manner( P 0. 05,P 0. 01). Conclution: The two weeks' s administration of aqueous extract of Fructus Psoraleae showed an effect on the endogenous metabolites in serum,its mechanism of pharmacological effects and toxicity effect may be relate to amino acid metabolism,phospholipid metabolism,and glycolysis.
