诱导热休克蛋白70对感染后肠易激综合征小鼠的影响 被引量:1

摘要 目的探讨诱导热休克蛋白70(HSP70)对感染后肠易激综合征(PI-IBS)小鼠的影响。方法将84只雌性C57BL/6小鼠随机分为对照组、PI-IBS组、诱导+PI-IBS组、诱导组,每组21只。PI-IBS组给予旋毛虫感染;诱导+PI-IBS组先给予热预处理,再给予旋毛虫感染;诱导组只给予热预处理,正常对照组不给予处理。Western blot检测肠道HSP70蛋白表达量,观察肠组织病理学表现,对肠组织行炎症评分,观察腹壁撤退反射(AWR)以评估内脏敏感性,观察肠道传输时间及粪便Bristol评分以评估肠道动力,ELISA检测肠道炎症细胞因子浓度。结果 PI-IBS组、诱导组HSP70表达量较正常对照组明显升高(P<0.01),诱导+PI-IB组较PI-IBS组亦明显升高(P<0.01)。PI-IBS组肠道炎症评分明显高于正常对照组(P<0.01),而诱导+PI-IBS组明显低于PI-IBS组(P<0.01),诱导组与正常对照组差异无统计学意义(P>0.05)。PI-IBS组AWR评分明显高于正常对照组(P<0.01),诱导+PI-IBS组明显低于PI-IBS组(P<0.01),诱导组与正常对照组差异无统计学意义(P>0.05)。PI-IBS组肠道传输时间比正常对照组明显缩短(P<0.01),而诱导+PI-IBS组明显长于PI-IBS组(P<0.01),诱导组与正常对照组差异无统计学意义(P>0.05);PI-IBS组Bristol评分比正常对照组明显升高(P<0.01),而诱导+PI-IBS组明显低于PI-IBS组(P<0.01),诱导组与正常对照组差异无统计学意义(P>0.05)。与正常对照组比较,PI-IBS组IL-10浓度明显降低(P<0.01),而IL-17、IFN-γ、TNF-α浓度均明显升高(P<0.01);与PI-IBS组比较,诱导+PI-IB组IL-10浓度明显升高(P<0.01),而IL-17、IFN-γ、TNF-α浓度均明显降低(P<0.01);诱导组与正常对照组IL-10、IL-17、IFN-γ、TNF-α浓度差异均无统计学意义(P>0.05)。结论诱导肠道HSP70可通过调节肠道炎症细胞因子平衡来改善PI-IBS时的炎症状态。 Objective To investigate the effect of the pre- induced intestinal heat shock protein 70( HSP70) on mice with post- infectious irritable bowel syndrome( PI- IBS). Methods 84 female C57 BL /6 mice were randomly divided into four groups: the control group,PI- IBS group,induction + PI- IBS group and induction group,with 21 mice in each group. The mice in the PI- IBS group were infected in vivo with Trichinella spiralis. The mice in the induction +PI- IBS group were given heat pretreatment before infection. The mice in the induction group were only given heat pretreatment without infection. The mice in the control group were given no treatment. HSP70 expression in intestinal tissues was measured by Western blot. Terminal ileum tissue was processed for pathological examination. Inflammation score of intestinal tissue was documented. Abdominal withdrawal reflex( AWR) was used to assess the visceral sensitivity. Colon transportation time and Bristol score were used to assess intestinal motility. Concentrations of intestinal pro- inflammatory cytokines were detected by ELISA. Results HSP70 expression in the PI- IBS group and induction group were higher than that in the control( P〈0. 01),while induction + PI- IBS group was higher than PI- IBS group( P〈0. 01). Inflammation score of intestinal tissue in the PI- IBS group was higher than that in the control group and induction + PI-IBS group( P〈0. 01),while there was no significant difference between the induction group and control group( P〉0. 05). AWR score of the PI- IBS group was higher than that of the control group and induction + PI- IBS group( P〈0. 01),while there was no significant difference between the induction group and the control group( P〉0. 05). Colon transportation time of the PI- IBS group was shorter than that of the control group and induction + PI- IBS group( P〈0. 01),while there was no significant difference between the induction group and the control group( P〉0. 05).
