摘要
目的观察经验方复荣通脉胶囊对糖尿病大鼠心肌转化生长因子β1(TGF-β1)、结缔组织生长因子(CTGF)的影响,探讨其对糖尿病心肌病的可能作用机制,为临床防治糖尿病心肌病提供可靠的实验依据。方法将雄性Wistar大鼠88只随机分为正常组、模型组、复荣通脉胶囊低剂量组及复荣通脉胶囊高剂量组,每组22只。除正常组外,其余组均采用腹腔注射链脲佐菌素(STZ)的方法建立糖尿病大鼠模型。造模成功后,复荣通脉胶囊低剂量组和高剂量组分别给予复荣通脉胶囊混悬液0.67 g/(kg·d)和1.34 g/(kg·d)灌胃,正常组和模型组给予等体积蒸馏水灌胃。连续灌胃8周后,测量各组体质量、血糖、TC、TG,免疫组化法检测心肌组织中TGF-β1、CTGF表达情况。结果灌胃8周后,正常组体质量明显高于灌胃前(P<0.05),模型组和复荣通脉胶囊低剂量组、高剂量组体质量均明显低于灌胃前及正常组(P均<0.05);各组血糖水平与灌胃前比较差异均无统计学意义(P均>0.05),模型组和复荣通脉胶囊低剂量组、高剂量组比较差异也均无统计学意义(P均>0.05);各组间TC比较差异均无统计学意义(P均>0.05),模型组TG明显高于正常组(P<0.05),复荣通脉胶囊低剂量组、高剂量组TG与模型组比较差异均无统计学意义(P均>0.05);复荣通脉胶囊高剂量组心肌组织中TGF-β1、CTGF表达水平明显低于模型组和复荣通脉胶囊低剂量组(P均<0.05)。结论复荣通脉胶囊可下调TGF-β1、CTGF在糖尿病大鼠心肌组织中的表达,从而发挥其对糖尿病心肌病的治疗作用,但具有剂量依赖性。
Objective It is to observe the intervention effects of Furongtongmai capsule on the expression level of myocardial TGF-β1and connective tissue growth factor( CTGF) in diabetic rats,and to explore its possible mechanism on diabetic cardiomyopathy. Methods 88 male Wistar rats were randomly divided into four groups: normal control group( NC group),diabetic model control group( DM group),Furongtongmai capsule low-dose( FRL) and high-dose treated( FRH) groups with 22 rats in each group. The diabetic rat models were created by intraperitoneal injection of streptozocin( STZ) in all groups except NC group. After the models were successfully established,FRL group and FRH group were given Furongtongmai capsule 0. 67 g /( kg·d) and 1. 34 g /( kg·d) respectively by lavage,NC group and DM group were given normal saline by lavage. After treatment for 8 weeks,body weight( BW) was measured,blood glucose( BG),TC and TG were tested with automatic biochemical analyzer,and myocardial TGF-β1and CTGF were detected by immunoshistochemistry assay. Results After 8 weeks,BW of the rats were higher in NC group while that in DM group and FRL and FRH groups were lower than those before lavage( P0. 05). No significant change of BG was found in every group compared with that before lavage( P0. 05). There was no significant difference in TC and BG among every group( P0. 05),but the level of TG was higher in DM group than that in NC group( P 0. 05),no significant difference in TG was found between Furongtongmai capsule treatment groups and DM group( P0. 05). The expression level of TGF-β1and CTGF in FRH group was lower than that in DM group and FRL group( P0. 05). Conclusion Furongtongmai capsule can down-regulate the expression of TGF-β1and CTGF in myocardial tissue in DM rats,thus to play its treatment effect on diabetic cardiomyopathy,but dose dependence is also needed.
