摘要
目的:探讨低剂量60Coγ射线照射诱导人永生化淋巴细胞AHH-1中P53结合蛋白1(53BP1)焦点水平与照射剂量之间的关系。方法:用^(60)Coγ射线照射AHH-1细胞,照射剂量分别为0 m Gy、10 m Gy、25 m Gy、50 m Gy、100和200 m Gy,吸收剂量率为20 m Gy/min,37℃修复30 min后进行免疫荧光染色,利用激光共聚焦扫描显微镜分析细胞中53BP1焦点形成情况。结果:在剂量为0~200 mG y的^(60)Coγ射线照射30 min后,AHH-1细胞中53BP1焦点在每个细胞中的数目为0~4个,均符合泊松分布。从50 m Gy照射剂量开始53BP1焦点水平显著性增加,且焦点水平与照射剂量具有剂量-效应关系,剂量-效应曲线为y=(0.03×10^(-2))x+2.32×10^(-2)(R^2=0.9236,P<0.01)。结论:在低剂量电离辐射作用下,53BP1焦点水平具有较好的剂量-效应关系。
Objective: To investigate the dose- response relationship between the levels of 53BP1 foci in the lymphocyte cells AHH-1 and the absorbed doses of low dose irradiation. Methods: Lymphocyte cells AHH-1 were exposed to different absorbed dose levels of 60Co v-rays (0, 10, 25, 50, 100 and 200 mGy at the absorbed dose-rate of 20 mGy/min). Immunofluorescence analysis was carried out 50 min after irradiation and the formation of 53BP1 foci (foci/per cell) was analyzed using laser scanning confocal fluorescence microscope. Results: Within the dose range of 0-200 mGy, the levels of 53BP1 foci were conformed to the Poisson distribution. The levels of 55BP1 were enhanced when the dose level was above 50 mGy, and the dose-response curve was 2 JJ ~ well fitted as y= (0.03X l02)x+2.32X 10- (R2=0.9236, P〈0.01). Conclusion: There is a good dose-response relationship between the levels of 53BP1 foci and the low doses, and hence it could be applicable as a biomarker of low dose radiation.
