焦油、尼古丁和瑞舒伐他汀对人脐静脉内皮细胞表达内皮细胞蛋白C受体的影响 被引量:1

Effects of tar, nicotine and rosuvastatin on expression of EPCR in human umbilical vein endothelial cells

摘要 目的探讨焦油、尼古丁和瑞舒伐他汀对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)表达内皮细胞蛋白C受体(endothelial cell protein C receptor,EPCR)的影响。方法体外分离培养HUVECs并传代至4~6代,将其分为对照组,50 mg/L焦油组,10^(-4) mol/L尼古丁组,10^(-5) mol/L瑞舒伐他汀组。各组分别孵育6 h和24 h后,收集细胞,使用流式细胞术和实时荧光定量聚合酶链式反应(quantitative real time polymerase chain reaction,q RT-PCR)分别检测并比较细胞EPCR蛋白表达和胞浆中信使核糖核酸(messenger ribonucleic acid,m RNA)水平。结果 (1)与同时间点对照组比较,EPCR蛋白表达在焦油组孵育6 h(t=29.513,P<0.001)及24 h(t=41.759,P<0.001)后均明显上调,差异有统计学意义;而尼古丁组、瑞舒伐他汀组均无明显变化。(2)与同时间点对照组相比,m RNA表达量在焦油组孵育6 h(t=20.753,P<0.001)和24 h(t=19.897,P<0.001)后均明显增加;而尼古丁组、瑞舒伐他汀组均无明显变化。结论焦油可上调HUVECs表面EPCR的表达,而尼古丁和瑞舒伐他汀对EPCR的表达无影响。 Objective To explore the effects of tar, nicotine and rosuvastatin on expression of endothelial cell protein C receptor (EPCR) in human umbilical vein endothelial cells (HUVECs). Methods HUVECs were isolated and cultured in vitro, and were randomly divided into control group, 50 mg/L tar group, 10.4 mol/L nicotine group, 10.5 mol/L rosuvastatin at passage 4 to 6. All groups were incubated for 6 h and 24 h, then collected the HUVECs. Flow cytometry and quantitative real time polymerase chain reaction (qRT-PCR) were respectively used to detect and compare expression of EPCR protein and the EPCR level of messenger ribonucleie acid (mRNA) of in each HUVECs group. Results (1) Compared with control group at the same time point, expression of EPCR protein in 50 mg/L tar group after incubated for 6 h (t=29.513, P〈0.001) and 24 h (t=41.759, P〈0.O01) were both increased, and the differences were statistically significant; but in 10-4 mol/L nicotine group and l0"5 mol/L rosuvastatin group had no obvious change. (2) Compared with control group at the same time point, expression of EPCR mRNA in 50 mg/L tar group after incubated for 6 h (t=20.753, P〈0.001) and 24 h (t=19.897, P〈0.001) were significantly increased; but there were no significant change in 104 mol/L nicotine group and 104 mol/L rosuvastatin group. Conclusions Tar has the effect of up-regulating the expression of EPCR in HUVECs, whereas nicotine and rosuvastatin have no effect on the expression of EPCR in HUVECs.
